Objective Malignant tumor diseases threat human's health seriously,there are many ways to treat them at present,For example: surgical operation,radiation therapy,chemical treatment,Gene therapy.Now,there are many chemotherapeutic drugs used in clinical practice,Their finalities are not clear.They kill the tumor cells,But the normal cells are also been killed.They Cause great harm to human body.Search a antitumor drug with clear effects and high efficiency,at the same time with low toxicity.To reduce the harm to the body to the greatest extent become the direction of antitumor drugs' studies.According to the report,the onset of gastric cancer on a globle scale is relatively common.All of the malignant tumor,the incidence of gastric cancer is number four.In 2013,the new happens of gastric cancer in USA are 21600 cases.10990 people died of gastric cancer.There are many kinds of tumors in the digestive tract,malignant tumors are common.Among these malignant tumors,The most common one is gastric cancer,gastric cancer's incidence is also the highest.The incidence of gastric cancer is very different between men and women.Men's probability of suffering from gastric cancer is double that of women.The age of patients with gastric cancer has a wide distribution.The probability increase obviously when the age reaches more than fifty years old.Most patients get into the middle-late when they find their gastric cancer.Cancer cells have moved to other places.Even intruded into other organs and tissues.The best time for surgery was missed.Many scholars at home and abroad attache great importance to the drug therapy for gastric cancer.There are many reports about gallic acid.Many scholars at home and abroad have studied gallic acid.According to the current report,gallic acid is a kind of organic acids.Its English name is gallic acid.GA for short.There is abundant of gallic acid in the cornus.In addition to this,other Chinese herbal medicine also has abundant of gallic acid.For example,Tibetan medicine rhodiola.In recent years,correlational research found that,gallic acid has a certain potential for the treat of various diseases.Especially the treatment of tumor received extensive attention of scholars at home and abroad.GA can block the growth of tumor.It makes little difference to normal cells when acts on the cancer cells.GA may be developed into a new cancer drugs applied to clinical practice.In this research,by discuss gallic acid's effect on the gastric cancer cell line SGC7901's biological behavior,and themechanism of these influences,to guide the clinical treatment of gastric cancer.Methods This experimental makes the gastric cancer cell line SGC7901 as research object.After the cells cultured,choosing logarithmic phase cells for the experimental.Testing gallic acid's influence on the proliferation of gastric cancer cell by MTT method.Get the effective concentration of gallic acid to cell proliferation;Testing gallic acid's influence on the invasive ability of gastric cancer cells by transwell;Testing change of the cell cycle and apoptosis by Flow Cytometer;Testing the express of cyclin D1 protein by Western Blot.Result 1.Testing gallic acid's influence on the proliferation of gastric cancer cell by MTT method.After 24 hours use of GA,making the the concentration of GA as 1.6?3.2?6.4?12.8?25.6 ?g/m L.The inhibition rate of GA to gastric cancer cell : 13.3%?20.5%?36.6%?48.7%?49.9%(p<0.05).After 48 hours use of GA,making the the concentration of GA as1.6?3.2?6.4?12.8?25.6 ?g/m L.The inhibition rate of GA to gastric cancer cell : 18.9 %?29.6% ? 47.7% ? 68.8% ? 70.2%(p < 0.05);After 72 hours use of GA,making the the concentration of GA as 1.6?3.2?6.4?12.8?25.6 ?g/m L.The inhibition rate of GA to gastric cancer cell : 33.1 %?44.3%?67.9%?76.8%?77.8%(p<0.05).2.Testing gallic acid's influence on the invasive ability of gastric cancer cells by transwell.Making the the concentration of GA as 0 ? 6.4 ? 12.8 ?g/m L.The number of migratory cells: 139.25±20.55,84.75±19.94,59.00±8.91.To compared with control groups,the difference is statistically significant(F=22.406,P=0.000).3.Testing change of the cell cycle by Flow Cytometer.The result: the cell number of gastric cancer Is proportional to the concentration of GA.Making the the concentration of GA as 0?6.4?12.8 ?g/m L.The Cell proportion of G0 / G1 phase: 56.84%?62.58%?68.03%.The trend is increased.The difference is statistically significant(P<0.05).4.Testing change of the cell apoptosis by Flow Cytometer.The result: Early apoptosis cells number has positive relationship with the concentration of GA.Making the the concentration of GA as 0?6.4?12.8 ?g/m L.The early apoptosis rate of gastric cancer cell:2.54%?3.54%?10.22%.The difference is clear(P<0.05).5.Testing the express of cyclin D1 protein by Western Blot.Making the the concentration of GA as 0?6.4?12.8 ?g/m L.Compared with the blank control groups,the relative expression of Cyclin D1: 1.05±0.04,0.96±0.04,0.75±0.22.Using ImageJ analysesgray level,after the treatment of gallic acid,the expression of Cyclin D1 decreased obviously.The difference is statistically significant(F=90.315,P=0.000).Conclusion The proliferation,migration,and invasion of gastric cancer cell line SGC7901 can be blocked by GA;GA has a promoting effect on the cell apoptosis;It can block the cell cycle.The mechanism isthat,GA can hindered the Cyclin D1 gene protein expression.
