Paeonia rockii is a perennial woody plant of Ranunculaceae?Paeonia?,which is the special herb and treasured flower of China western region.Its root bark was used for a common tradional Chinese medicine recorded in Chinese materia medica standards of Gansu province,which has the efficacy of clearing heat and cooling blood,invigorating blood circulation and eliminating blood stasis.In order to develop and utilize the Paeonia rockii,this paper makes a overeview on relevant research of the food and medicine value of flower,seeds,root,pollen and leaf of Paeonia.On this basis,a series of studies about the quality standard and antioxidant activity of Paeonia rockii pollen have been done.The major research contents and results are as follows:1.The preliminary study on the quality standard of Paeonia rockii PollenAccording to general principles method of Chinese Pharmacopoeia?2015 edition,Volume 4?,the character and microscopic traits of Paeonia rockii pollen were described,and the total ash,acid insoluble ash,extract from Paeonia rockii pollen were analyzed in this study.The identification of the quercetin,luteolin kaempferol and isorhamnetin from Paeonia rockii pollen were performed by TLC.The content of total flavonoids were determined by UV-visible spectrophotometry.The result showed that the Paeonia rockii pollen was yellow powder with body light,faint and bitter.Under microscope,the shape of pollen particle was analogous round or oval,diameter was 32-38?m,which had reticulate sculpture on its surface with three quasi-holeditc.The content of ash,acid insoluble ash,extract of the Paeonia rockii pollen were 7.45%,1.55% and 28.99%,respectively.Its TLC identification had good reproducibility with clear spots;the TLC method was suitable for qualitative analysis of Paeonia rockii pollen.The average content of total flavonoids in Paeonia rockii pollen was 15.80mg·g-1.2.Optimize the extraction process of total flavonoids in Paeonia rockii pollen by response surface methodologyAgilent Eclipse plus C18 column?250mm×4.6mm,5?m?was adopted.The mobilephase was methanol-acetonitrile-0.1% phosphoric acid?25:13:62?at the flow rate of1.0mL· min-1.The detection wavelength was 360 nm and the column temperature was35 ?.then the method of simultaneous determination of quercetin,luteolin and isorhamnetin in pollen of Paeonia rockii by RP-HPLC is builded.Based on single factor experiments to select the extraction conditions for total flavonoids from Paeonia rockii pollen.The comprehensive scores of content of total flavonoids and total contents of three flavonoids?luteolin,quercetin,isorhamnetin?as indexes.The Box-Behnken test design were used to optimize 3 factors,which were ethanol concentration,solid-liquid ratio and extraction time.Response surface methodology was used to optimize the extraction technology of total flavonoids from Paeonia rockii pollen.The result show that the optimal parameters were as follows: Reflux extraction 2 times,73% as ethanol concentration,1:22 g/mL as material-liquid ratio,100 min as extraction time.Under these conditions,the observed value and predicted values was very close,and the deviation was 1.60%.The optimized extraction technology of total flavonoids from Paeonia rockii pollen is feasible.3.Study on the antioxidant activity of different concentration alcohol extracts of Paeonia rockii pollenThe antioxidant activity of different concentration alcohol extracts of paeonia rockii pollen were evaluated by cupric reducing power method,DPPH radical scavenging activity method and ABTS radical scavenging activity method.The result show that: The amount of antioxidant activity of gallic acid of anhydrous ethanol extract,75% alcohol extract,50% alcohol extract,25% alcohol extract,water extract from paeonia rockii pollen were 26.00,28.33,28.90,14.98,9.24 mg/g,respectively.The sequence of the ability of scavenging DPPH free radicals and ABTS radical were Vc>50% alcohol extract > 75% alcohol extract >anhydrous ethanol extract> 25%alcohol extract > water extract.The different concentration alcohol extracts of paeonia rockii pollen has certain antioxidant activity,especially for 50% alcohol extract and 75% alcohol extract.
