Objective: To explore the expression of GATA-1 and GATA-2 in bone marrow of patients with high-altitude polycythemia(HAPC).Methods: Bone marrow were extracted from 15 patients with HAPC and 10 patients without hematology disease.HAPC were determined by diagnostic criteria for the sixth international plateau medicine and hypoxia physiology conference,while hematological disease were eliminated from these 10 patients.Myeloid CD235+ cell were collected by density gradient centrifugation and magnetic cell sorting.The expression of GATA-1 and GATA-2 were detected by RT-PCR and western blot respectively.Results: The expression of GATA-1 mRNA in CD235a+ cell of HAPC bone marrow(HAPC group)was higher than those in CD235a+ cell of bone marrow discharged from hematopoietic disease(Control group)(P<0.05),which was11145371096.46±8925.35,4825140.79±2401.09 respectively.Compared with control group,GATA-1 protein expression was higher in HAPC group(1.22±0.35 in HAPC group and 0.87±0.17 in control group)(P<0.05).GATA-2 mRNA was only expressed in 2 patients in HAPC group and 3 patients in Control group,and its protein expression can't be detected.Conclusion: The expression of GATA-1 in HAPC bone marrow CD235+ cell was higher than those in the control group,and the expression of GATA-2 may decrease along the erythropoiesis progress.The influence of GATA-1 on advanced hematopoietic cells may play a role in HAPC pathogenesis.
