Objective:To investigate the effects of tauroursodeoxycholic acid on modified A11 en s weight drop method to construct rat acute spinal cord injury model of neural cell autophagy.Methods:A total of 72 male SD rats,weighing 250-300 g,were selected for 6-8 weeks.They were randomly divided into three groups: sham operation group,operation group,control group(saline group),TUDCA group(high and low dose group of two).The animals were sacrificed at 24,5 and 10 days after spinal cord injury.According to BBB scores,injury of Rats after hindlimb motor function score and sudden death before;by means of electron microscope,HE staining,TUNEL assay,immunohistochemistry technique to observe cell necrosis and autophagy;using Western blotting method to detect the autophagy related factor(Beclin-1),the expression of apoptosis related factor caspase-3.Expression;and using the RT-PCR method were used to detect the protein mRNA.Results:In BBB group,the activity of rats in sham operated group was not limited.The c ontrol group and experimental group,control group,BBB score was significantly lower in the experimental group,to reflect the activity of the rats on the visible activity is slightly worse t han the level of the experimental group in the experimental group of drug dose effects on rat a ctivity had no significant difference.The physiological function of the saline group was significantly reduced by elec tron microscopy,and the occurrence of autophagy was observed.Biological characteristics of injured spinal cord neurons after TUDCA intervention;.Comparison of the differences in the morphology of spinal cord under light micro scope with HE staining.The normal spinal cord morphology,spinal cord neurons and glial cel ls were intact,and there were no obvious necrosis cells.The control group of spinal cord injur y,can see cell vacuolar degeneration,edema and hemorrhage in large cells,loose organization al structure is obvious,gray tissue destruction,accompanied by a large number of neurons and neuron necrosis,cell survival,part of the nuclear pyknosis of the spinal cord injury.The experimental group was treated,compared with the control group were the same as hemorrhage,edema and necrosis is the main performance,but the control characteristics of the largest group in the section near the center of the injury,the same damage,can see damage to expand the scope of the adjacent nerve cells damaged significantly,some cells had the morphology of T the obvious change in the experimental group,although the range of cell damage than before also increased,but compared with the control group,and the range is smaller,and the extent of damage of adjacent nerve cells with light.Western blotting method was used to detect the expression of autophagy related factor(Beclin-1)in each group.The expression of Beclin-1 in experimental group were higher than the control group,and there were significant differences between the TUDCA,the same to the dose level is no significant difference of the Western blotting method,detection of apoptosis related factor(caspase-3)expression in each group.The expression of Caspase-3 at different time points in the SCI control group was significantly higher than that in the sham operation group,while the expression of Caspase-3 in the experimental group was lower than that in the control group after the intervention of TUDCA.However,there was no significant difference in the high and low dose of TUDCA in the experimental group.Conclusion:The intervention of TUDCA can significantly active ate the expression of autophagy in rats with acute spinal cord injury and inhibit the apoptosis of nerve cells.
