Purpose:The aims of this study are to detect the expression levels of long non-coding RNA(lnc RNA)H19 in patients with non-small cell lung cancer(NSCLC),and to explore the relationship of lnc RNA H19 with clinicopathologic factors,tumor angiogenesis,and prognosis.Methods:1.Real-time quantitative polymerase chain reaction(q RT-PCR)analyses were utilized to investigate H19 expression status in both tumor and normal tissues of 130 patients with NSCLC.And we explore the relationship between lnc RNA H19 and clinicopathologic factors.2.Intratumoral microvessel density(MVD)was recorded by counting CD31-positive immunostained endothelial cells to evaluate the tumor angiogenesis status in non-small cell lung cancer.And we explore the relationship of MVD with clinicopathologic factors and lnc RNA H19.3.According to postoperative follow-up results,we analyse the relationship of lnc RNA H19 and MVD with prognosis.Results:1.H19 expression level in tumors was significantly higher than those in the corresponding normal tissues in the patients with NSCLC(p<0.01).Upregulation of lnc RNA H19 was significantly associated with tumor-node-metastasis(TNM)stage(p<0.001),lymph node metastasis status(p=0.014)and tumor size(p<0.001).2.High MVD was significantly associated with TNM stage(p=0.001),lymph node metastasis status(p=0.021).3.Upregulation of lnc RNA H19 was detected and was significantly associated with MVD(p<0.001).4.Patients with high expression level of H19 had significantly poor overall survival(OS)(38.2% versus 89.1%,p<0.001)and disease-free survival(DFS)(32.2% versus 86.0%,p<0.001)at 3 years after operation.5.Patients with high MVD had significantly poor overall survival(OS)(36.6% versus 89.3%,p<0.001)and disease-free survival(DFS)(28.9% versus 87.8%,p<0.001)at 3 years after operation.6.Upregulation of H19 was an independent prognostic factor for unfavorable OS(p=0.001)and DFS(p<0.001).Meanwhile,high MVD was also an independent prognostic factor for unfavorable OS(p=0.001)and DFS(p<0.001).Conclusions:1.Upregulation of lnc RNA H19 was common in NSCLC.There was significant correlation between H19 expression and TNM stage,lymph node metastasis status and tumor size in NSCLC.2.High MVD was significantly associated with TNM stage,lymph node metastasis status in NSCLC.3.There was significant correlation between upregulation of lnc RNA H19 and tumor angiogenesis.4.Increased expression level of H19 and high MVD associated with poor prognosis in NSCLC.5.Upregulation of H19 and high MVD were independent prognostic factors for unfavorable OS and DFS in NSCLC.
