Research On Processing Of Recipe Ginseng Agent And Its Anti-hepatoma Effect

Tumor prevention is a worldwide problem with the extremely high incidence of the disease,liver cancer.Liver cancer's characteristics are the concealment onset,late discovery,low cure rate,the high fatality rate which have a rising trend year by year.The research of traditional Chinese medicine(TCM)to cure cancer has become a hot spot currently,and they focus on the single TCM research only,Panax ginseng,Antler plate,Ophiopogonis radix,Astragalus membranaceus that all have antitumor effect,but the matching with four drugs and its antitumor effect on H22 have not been reported.The experiment is compatibility of the four kinds of TCM into the recipe ginseng agent.Firstly,we optimized technology of recipe ginseng agent based on ginsenosides;Secondly,we processed optimization of recipe ginseng agent on the basis of sensory score;Thirdly,the contents of ginsenosides,polysaccharide and amino acid were detected by HPLC and phenol-sulfuric acid in order to make quality standards of recipe ginseng agent;Finally,the author confirmed the antitumor activity,mechanism and safety evaluation of recipe ginseng agent.Experimental results were as follows:1.The best extraction processing of the recipe ginseng agent: temperature was 100 ?,solid-liquid ration was 1:31,boiled times was 160 min.2.The best forming processing of recipe ginseng agent: compound ginseng's content was 15g/50 mL,fructose's content was 24.9g/50 mL,carrageenan's content was 1.5g/50 mL,the formula of the product had claybank color,uniform color,plumpness,great taste,moderate sweetness,good chew.3.15 kinds of ginsenosides and 16 kinds of amino acids were detected from recipe ginseng agent.As calculation with the ginseng single herb,the contents of ginsenosides were significantly higher than those of single herb in recipe ginseng agent(P<0.05),which can improve the rare ginsenosides contents such as Rg2,Rh4,Rg3,Rg5,CK(P<0.05)and decline the four kinds of ginsenosides contents such as Rg1?Re?Rb1?Rb2?Rc?Rd(P<0.05),the contents of polysaccharides,essential amino acid,medicinal amino acid and total amino acid in recipe ginseng agent were significantly higher than those of single herbs(P<0.05).As calculation with the ginseng single herb,the essential amino acid,medicinal amino acid and total amino acid contents were 19.66%,44.01% and 73.85% respectively in recipe ginseng agent which was higher than the ginseng herb(P<0.05);as calculation with the antler plate single herb,the medicinal amino acid,essential amino acid and total amino acid contents were 9.83%,22.01%,36.93% respectively in recipe ginseng agent which was higher than the antler plate herb(P<0.05).4.The high dose of recipe ginseng agent could inhibit the tumor growth of H22 tumor-bearing mice,increase the spleen index and thymus index(P<0.05),accelerate the early apoptosis rate and total apoptosis rate of tumor cell(P < 0.05);improve the immunoglobulin contents such as IgG,IgE,IgM(P<0.05),increase the levels of cytokine such as IL-2?IL-6?IL-12?IFN-??TNF-?(P<0.05),reduce the content of TGF-?(P<0.05).Compared with model group,the tumor tissue of the mice in various administration groups was destroyed,the cells were sparse and irregular,and the tumor presented necrotic lesions;the morphology of spleen cells was normal with discernible fringe,and the lymphocytes arranged densely.These results proved that recipe ginseng agent had certain protective effect on the immune organs,but spleen tissue destruction in 5-FU group was serious.recipe ginseng agent has improved the body immunity,so as to play its antitumor effect.5.Recipe ginseng agent's was safe and reliable.
