The context was tested and verified the monosaccharide composition of polysaccharide?GI-A,GI-U,GI-T and GI-S?from fruiting bodies of Gloeostereum incamatum,and investigated their anti-tumor effects on tumor-bearing mice with H22.Furthermore,it's studied the anti-tumor mechanisms of urea soluble polysaccharides of Gloeostereum incamatum.The purpose is to provide reference for research and drug development of Gloeostereum incamatum.1.The crude polysaccharides,GI-A,GI-U,GI-T and GI-S,were obtained from the different extractions that were extracted with pH=2 hydrochloric acid solution,6% urea solution,3% of the three trichloroacetic acid solution and 0.5M solution of sodium hydroxide.The protein of the polysaccharides was removed with Sevage method,after that,the active carbon was used to decolorization to obtain pure polysaccharides of Gloeostereum incarnatum : GI-A,GI-U,GI-T and GI-S.2.Method Establishment for determination of monosaccharide composition of Gloeostereum incarnatum polysaccharide Using aldoononitrile acetate gas chromatography to determine the monosaccharide composition of polysaccharide.The experimental results showed that the polysaccharides of Gloeostereum incamatum were composed of xylose,mannose and glucose.Their molar ratios are respectively: 1:1.2:1.4,2:2.9:1,1.5:1:2.1,1:1.4:2.2.3.Antitumor activity analysis of polysaccharides of Gloeostereum incamatum A series of indexes,including tumor weight,tumor inhibition rate,thymus index,spleen index,AST,ALT,BUN,CRE,TNF-?,IL-2,tumor tissue and spleen H&E staining,immunohistochemistry,were used to investigated the antitumor activity of polysaccharides.The results showed that:?1?The data of anti-tumor effects of tested groups with intragastric administration of polysaccharides of Gloeostereum incamatum,showed that polysaccharides of Gloeostereum incamatum could inhibit the H22 tumor growth in a certain degree.Among them,the GI-U group at high dose showed the most anti-tumor activity,and the anti-tumor inhibitory was 60.69%,which was just lower than the positive group with 65.69%;and the medium and low dose group also showed good antitumor effect with inhibition rates of 45.83% and 39.04%,respectively,while the GI-A also possessed good anti-tumor effect,which could inhibit the tumor growth with rates of 53.24% and 58.94%,the anti-tumor effect is obvious.GI-T and GI-S also showed a certain anti-tumor effect,but the effect is not obvious,for their tumor growth inhibitory rates were correspondingly only 44.57% and 49.79% at high dose.?2?According to the results of investigation and comparison of each index from the four tested groups,GI-U showed the most antitumor activity,GI-A was lower,while the GI-T and GI-S were not obvious.Furthermore,the tumor tissues of GI-A and GI-U groups were studied with H&E staining.The results showed that GI-A and GI-U played the role in inhibiting tumor growth,through inducing tumor cell apoptosis.?3?The antitumor activity of GI-U was analyzed by immunohistochemistry and western.It was found that GI-U could regulate the expression of protein and then play the role of anti-tumor.
