Objective:Glioma is the most common tumor in intracranial tumors,histological classification for astrocytoma,less sudden glioma,etc.These cancers exhibit a relentless malignant progression characterized by widespread invasion throughout the brain.And the high incidence and recurrence rate,high mortality and low cure rate.According to the World Health Organization(WHO)classification,most of the I grade gliomas occur in children and most of the prognosis is good,but the II grade glioma has a median survival of 5-8 years,III grade astrocytoma the median survival time is about 3 years,IV grade glioma’s median survival time only have 12-18 months.Galangin is a Small lipid-soluble molecules natural flavonoids what extract in galangal rhizome and exists in Propolis,many studies show that it has antibacterial and antiviral effects in vitro,such as liver cancer,lung cancer and esophageal cancer.Wnt/ β-Catenin signaling pathway is a classic signal pathway,this signaling pathway has the important relation on the proliferation,apoptosis,invasion and migration of cells.And this pathway has intimate relation on the mitochondrial pathway of apoptosis.Research the effect of inhibiting proliferation and promote apoptosis on glioma cells about Galangin.This study was designed MTT test cell proliferation、using Hoechst 33342 staining observed apoptotic cells、flow cytometric method to detect apoptotic cells is described to determination of Galangin in U251 and U87 glioma cell proliferation and apoptosis.Western blot detection Galangin in glioma cells after Wnt/beta-Catenin signaling pathway and mitochondrial pathway related proteins table of change to determine Galangin in glioma cell apoptosis in vitro experiments and the role of affect migration and analyze its related mechanism.Methods:1、Determined the effect of the galangin inhibiting proliferation in U251 and U87 glioma cells by MTT method.2、Using Hoechst 33342 staining observed apoptotic cells about galangin induced U251 and U87 glioma cells apoptosis.3 、 Using flow cytometric method to detect apoptotic cells is described to determination of Galangin in U251 and U87 glioma cell proliferation and apoptosis in different concentrations under the action of Galangin.4、Using WESTERN BLOT technique to detect the expression of change about the Wnt/beta-Catenin signaling pathway mitochondrial apoptosis related proteins such as beta-Catenin,BCL-2,BAX,Cleaved caspase 3,Cleaved caspase 9,Cyt C,PARP.Results:This research reminder that,under the different time and density the effect of Galangin Inhibition the proliferation of glioma cells U87 and U251 is distinctly different,and presents a kind of relation with the amount of time and dose related effect,under the 100 μM、200 μM、300 μM 和 400 μM of Galangin the IC50 of U251 cells at the 12 h、24 h、48 h and 72 h is 281 μM、321 μM、276 μMand 229 μM,U87 is 289.4μM、261.1μM、247.4μM and 225.3μM.The apoptosis of glioma cells under the effect of Galangin are increase clearly,and the concentration and time are very important cause.We have a same change at the flow cytometric method to detect apoptotic cells is described.The expression of protein about Wnt/β-Catenin pathway and the apoptosis-related protein such as β-Catenin、BCL-2、BAX、Cleaved-caspase 3、Cleaved-caspase 9、Cyt C 、PARP have obvious change under the effect of Galangin,and the results have statistical significance.Conclusion:In the research we find that,Galangin can inhibit the proliferation of glioma cells U251、U87 obviously,and induce U251、U87 cells apoptosis.The expression of Wnt/β-Catenin and mitochondrial pathway protein is different about the effect of different concentrations of Galangin in the glioma cell U251 and U87.So we determine that Galangin can inhibit the proliferation of glioma cells and can promote the glioma cell apoptosis,this effect is closely relateda bout the Wnt/β-catenin signaling pathway.
