| Objective To observe whether Nitrosyl(HNO)have a protective effect on the cardiac function of rats having heart failure and discuss the possible mechanism,The rats with heart failure were intervened by nitrosyl hydrogen donor.Methods 50 male Wistar rats were selected and 42 rats out of them were selected to establish heart failure model by a ligation of anterior descending branch,18 rats survived after the surgery,which were randomly divided into heart failure group((HF group n=9)and Angeli's salt treatment group(AS group n=9),the remaining 8 rats were taken as a sham operation group(Sham group n=8).the AS group rats were treated with HNO donor Angeli's salt by intraperitoneal injection of 1 mg/kg/d,and the rats in the HF group and the Sham group were given the same amount of normal saline.After the intervention with 2 weeks,2rats died in the HF group and 1 died in the AS group.The remaining rat's survival states were in poor condition and then the experiment was terminated.To measured the Serum NT-ProBNP levels and cardiac ultrasonography,the measurement parameters that contain the left ventricular end diastolic diameter(LVEDd),Left ventricular end-diastolic diameter(LVEDs),left ventricular ejection fraction(LVEF),the left ventricular short axis shrinkage-rate were measured(LVFS).Left ventricular systolic pressure(LVSP)Left ventricular systolic pressure(LVSP),left ventricular end diastolic pressure(LVEDP),maximal rate in the left ventricular pressure(+dp/dt)and maximal decrease in left ventricular pressure(-dp/dt)were measured by arterial catheters.The Western blot was used to detect the expression of sarcoplasmic reticulum calcium pump(SERCA2a)protein in the left ventricle non-infarcted area,to detect the SERCA2 a protein activity by the biochemical kit finally.Results Before the drug intervention,LVEDd,LVEDs and NT-ProBNP were significantly increased(7.04 ± 0.42vs5.10 ± 0.32,5.59 ± 0.42vs2.92 ± 0.30,377.97 ± 20.47vs83.48 ±4.69,p<0.01),and LVEF,LVFS were significantly decreased(49.89 ± 2.85vs81.25 ±2.19,20.67 ± 1.50vs42.88 ± 2.42,p<0.01)in HF group compared with Sham group.There were no significant difference between HF group and AS group in LVEDd,LVEDs,LVEF,LVFS,and NT-ProBNP(7.09 ± 0.42vs7.04 ± 0.42,5.62 ± 0.43vs5.59 ±0.42,50.22± 2.64vs49.89 ± 2.85,20.89±1.17vs20.67 ±1.50,381.20 ±20.81vs377.97 ±20.47,p>0.05).After the drug intervention,Compared with Sham group,LVEDP was significantly increased(18.39 ± 1.43vs7.18 ± 0.91,p<0.01),meanwhile +dp/dt and-dp/dt(2044.14 ± 236.82vs3950.13 ± 264.29,1966.43 ± 225.43vs3842.25 ±241.07,p<0.01),the expression of SERCA2 a protein(0.28 ± 0.031vs0.49 ±0.039,p<0.01)and the activity(7.47 ± 1.32vs14.34 ± 1.35,p<0.01)was significantly decreased in HF group,but there was no significant difference in LVSP(132.86 ±8.93vs138.75 ± 9.77,p>0.05).Compared with HF group,LVEF,LVFS,+dp/dt,-dp/dt increased significantly(53.25±2.43vs48.43±3.51,22.38±1.51vs20.00±1.63,2412.00±357.35vs2044.14±236.82,2302.25±335.54vs1966.43±225.43,p<0.05),NT-ProBNP and LVEDP were significantly decreased(393.88±23.37vs437.53±24.12,15.48±1.23vs18.39±1.43,p<0.05),the SERCA2 a protein activity was significantly enhanced in AS group(9.23± 1.44vs7.47 ± 1.32,p<0.05),but there were no significant difference in LVEDd,LVEDs,LVSP,and SERCA2 a protein expression amount(7.28 ± 0.48vs7.46 ±0.55,5.65 ± 0.47vs5.98 ± 0.57,136.38 ± 9.52vs132.86 ± 8.93,0.29 ± 0.036vs0.28 ±0.031,p>0.05).Conclusion 1.Nitroxyl(HNO)is able to enhance the cardiac function of the rats with heart failure,reduce the level of NT-ProBNP in peripheral blood,improve the hemodynamics,exert a protective effect on the heart of rats.2.The expression and the activity of SERCA2 a protein was decreased in the heart failure rats.3.The HNO could increase SERCA2 a protein activity of the heart failure rats,however it had no significant influence on SERCA2 a protein expression. |
