OBJECTIVE: Exosomes have been shown to mediate the transport of micro RNAs(mi RNAs)and mi RNAs in exosomes have been shown to be potential tumor biomarkers for the early diagnosis of tumors in a variety of cancers And judge the prognosis.To investigate the relationship between the expression of mi RNAs in the exosomes of the patients with renal cell carcinoma and the exosomes of renal cancer patients.To find out whether serum exosomes mi RNAs can be used as tumor biomarkers for early diagnosis of renal cell carcinoma.Methods:First,we extracted 30 patients with renal clear cell carcinoma(postoperative histopathologically confirmed T1)and 30 healthy controls,followed by age and sex.(Mi R-210,mi R-224,mi R-452,mi R34a)were screened out according to the results of gene chip analysis of mi RNAs in renal clear cell carcinoma and the related literature search.Real-time quantitative PCR(Quantitative Real-time PCR)validation comparison.We found that mi R-210 was highly expressed in serum of renal cell carcinoma(P <0.05),while the expression of mi R-224,mi R-452 and mi R-34 a was not statistically significant.And then by the extraction of renal cancer and normal control serum exosomes for q RT-PCR detection,the results found that renal exoclinic mi R-210 is also high expression,the difference was statistically significant,and in the withdrawal of exosomes Serum mi R-210 expression was significantly reduced,so we believe that renal cancer patients with serum mi R-210 from renal cell carcinoma release,and through the transit of the esophagus stable in the peripheral circulatory system.(SN12-PM6,786-O)and the exosomes in the normal tissue cells of renal tubular epithelial cells(HK2)were re-tested according to the results of serum RTPCR.The expression of mi RNAs in serum,serum exosomes and renal carcinomas was significantly higher than that in normal exosomes by real-time quantitative PCR.Results: The mi R-210,mi R-224,mi R-452 and mi R34 a were highly expressed in renal cell carcinoma by RT-PCR in renal cell carcinoma and normal control,but only mi R-210 was found in renal cell carcinoma Serum and renal cell carcinoma were significantly higher in serum exosomes(p <0.05).And the expression of mi R-210 was significantly down-regulated in the serum suspension of exosomes.Mi R-214,mi R-144,mi R-155 and mi R-21 were successfully selected according to the results of serum RT-PCR.The results showed that mi R-210 and mi R-21 were expressed in renal cell carcinoma and the difference was statistically significant(p <0.05).Conclusion:1.Through the extraction and identification of exosomes,it is proved that not only the renal cell carcinoma cells and normal tissue cells(HK2)can also secrete exosomes.The expression of mi R-210 in peripheral blood of patients with renal cell carcinoma and renal cell carcinoma was significantly higher than that of normal group(p <0.05).In the serum of kidney cancer,mi R-210 may be derived from the release of renal cell carcinoma cells and the mi R-210 is stable in peripheral blood of patients with renal cell carcinoma.It is expected to be a tumor biomarker of early diagnosis of renal cell carcinoma.(P <0.05),which may be helpful in improving the early diagnosis and improvement of prognosis of renal cell carcinoma.The difference was statistically significant(p <0.05).
