Purpose: To investigate the expression of mi RNA-126 in clear cell renal cell carcinoma tissues and adjacent normal tissues, analyze the relationship between the pathologic grade and clinical stage of the expression of mi RNA-126 in cc RCC. To study the effects of mi RNA-126 in renal carcinoma cells.Methods: 1.For real-time q PCR,the expressions of mi RNA-126 were performed in 30 cases of cc RCC and normal adjacent tissues. The Correlation of the expressions of mi RNA-126 and the pathologic grade and clinical stage were researched.2.The expressions of mi RNA-126-5p were detected by real-time q PCR in 786-O cells and HK-2 cells. The 786-O cells were transfected by has-mir-126-5p using Lipofectamine RNAi MAX to up-regulate the expression of mi RNA-126-5p. To study the changes of cell proliferation, invasion ability and apoptosis by MTT, transwell invasion test and flow cytometry respectively.Results:1.The outcome was demonstrated by real-time PCR that the expression of mi RNA-126-5p in 30 cases of cc RCC and its adjacent normal tissues(15.49 times higher in average than the expression of cc RCC)showed significant difference(P<0.05).Grouped by the pathological class,the expression of mi RNA-126-5p was 36.63±5.38 in pathological classⅠ, and 19.96±7.28 in pathological class Ⅱ-Ⅲ, which had no significantly correlations(P>0.05).In terms of clinical stage, the expression of mi RNA-126-5p was37.05±6.38 in clinical stageⅠ, and 24.03±6.15 in clinical stageⅡ-Ⅲ, which had no significantly difference(P>0.05).2. The expressions of mi RNA-126-5p in 786-O cells were lower than in HK-2 cells. Divided to treatment, control and blank group, 786-O cells were transfected successfully by has-mir-126-5p. The expression of mi RNA-126-5p in treatment group cells(42.86±7.01) was significantly higher than the expression in control group(27.46±8.43)(P<0.05).After 48 hours transfection, the 786-O cell survival rate in treatment group(80.82±0.47) was lower than the cells survival rate in control group(98.87±0.27),has significantly difference(P<0.05). Transwell invasion experiment outcomes showed that the cells numbers in treatment group(33.89±2.80) was significantly lower than the cells numbers in control group(53.67±2.23)(P<0.05).The cell apoptosis assay showed that the cell apoptosis rate in treatment group was 22.56±1.13,while the cell apoptosis rate in control group was 7.20±0.26,which had significantly difference compared with control group(P<0.05).Conclusions:1. The expression of mi RNA-126-5p in cc RCC was lower than in its adjacent normal tissues. No significant correlations were observed between mi RNA-126-5p expression and clinical stage, pathological class from patient with cc RCC. The expression of mi RNA-126-5p in 786-O cells was lower than in HK-2 cells.2. The expression of mi RNA-126-5p in 786-O cells which were successfully transfected by has-mi RNA-126-5p was significantly up-regulated. After up-regulating mi RNA-126-5p, the cells proliferation, invasion ability were significantly suppressed.
