| Objective In this study,the model of chronic pelvic inflammatory disease(CPID)in rat was treated by Danggui-Shaoyao-San(DSS).To explore the influence on immunologic function of T lymphocyte and interleukin-4(IL-4),interleukin-10(IL-10),nitric oxide(NO),to provide clues for further study of the pathogenesis of CPID,and to study the pharmacological mechanism that DDS improve immune function.Methods Rat models of CPID were established by infecting mixed bacteria and mechanical damage,sixty female rats were randomized into the normal group,CPID model group,Jingangteng capsule group,and high-,medium-,and low-dose DSS group.After 14 days of treatment,the rat uterus was observed for histopathological changes;the percentage of amount of CD3,CD4,CD8,Th,Tc cell in peripheral blood of rats were observed using Flow Cytometry;the expressions of nitric oxide(NO),interleukin-4(IL-4)and interleukin-10(IL-10)in the blood serum of rats were determined.Results DSS at the 3 dose could reduce the inflammatory cells in the endometrium,promoted epithelial cell proliferation;Histopathological scores were shown:compared with the model group,high and medium doses of DSS significantly improved the above-mentioned pathological changes(P<0.05),reduced lymphocyte infiltration,alleviated tissue edema and congestion,most layers of the uterine wall structure and glandular structures tended to be normal,more obvious effects were improved with the increase of dose.Jingangteng capsule group also has a significant therapeutic effect(P < 0.05),but the improvement of lymphocyte depletion and infiltration not as obvious as that in high DSS group;High and medium doses of DSS significantly increased Th,CD4,CD3,Th/Tc,CD4/CD8,while markedly decreased Tc,CD8(P<0.05);high and medium doses of DSS obviously increased IL-4,IL-10 expression level,and markedly decreased NO expression level(P < 0.05);Compared with Jingangteng capsule group,the levels of IL-4 of rats in high-dose DSS group increased obviously(P < 0.01),and the levels of NO decreased dramatically(P < 0.05).Conclusion The research suggested that the model group have abnormality of immune cells and cytokines and various immune cells and cytokines involvein and develop into CPID,immunopathology may play an important role in the pathogenesis of CPID.Through DSS treatment,which can improve the immune state and reduce the inflammatory reaction,DSS can improve the immune function through multiple targets,its working mechanisms may be as follows:(1)It increase Th cells so that the immune tend to be stable and reduce Tc cells to relieve damage of tissue,ratio of Th to Tc restores to the normal level,cellular immunity recover the normal balance.(2)It increase anti-inflammatory cytokine levels so proportion of pro-inflammatory and anti-inflammatory cytokines achieves balance,effectively inhibits inflamed growth and promote tissue regeneration and repair.(3)It eliminates oxygen-derived free radical and NO in the body and reduces cytotoxic to protect normal cells from damage.(4)It regulates immune function with multiple levels ways in order to resist tissue adhesions. |
