Part 1 Study on the bile acid-absorbing capacity of hydrotalcite in vitroObjective To evaluate the bile acid-binding capacity of hydrotalcite at different p H in different doses in vitro.Methods Different doses of hydrotalcite were added to the diluted bile with p H values of 4,6,7,8 and 10 respectively,which were set to imitate the p H values of intestinal conditions.Total bile acid of the supernatant fluid was mensurated after reaction,and the bile acid-absorbing rate of hydrotalcite was calculated.Results1.There was no cross correlation between p H value and doses of hydrotalcite in the absorbing of total bile acid.The relation of total bile acid-absorbing rate with p H value and doses of hydrotalcite was AR=0.442-0.008*p H+1.425D(F=55.354,P<0.001,R=0.748).2.Hydrotalcite can bind bile acid both in acidic and alkaline conditions.But the bile acid-absorbing rate of hydrotalcite in acidic conditions was higher.3.If the concentrations of total bile acid of solution were limited,the total bile acid-absorbing rate was positively correlated with the doses of hydrotalcite.Conclusions Hydrotalcite can bind bile acid both in acidic and alkaline conditions.And there were positive correlations between bile acid-absorbing rate of hydrotalcite and doses of hydrotalcite.Part 2 Effects of hydrotalcite on the treatment of diarrhea induced by bile acid on ratsObjectives To imitate the distribution of bile acid in the intestinal conditions of patients with bile acid diarrhea,to assess the influence of bile or bile acid on the fecal water content of rats,and to explore whether the fecal water content can be alleviated on the treatment of hydrotalcite.Methods Male SD rats were randomly allocated into four groups: diluted human bile group(n=6),CA group(n=9),CDCA group(n=9)and CDCA with CA group(n=9).Feces were collected before and after the treatment with bile acid and hydrotalcite,respectively.Consistence of stool in different conditions was observed,then the weight of feces were mensurated before and after drying.The fecal water content was calculated,and concentrations of total bile acid were mensurated in feces of some rats.Results1.Diluted human bile could lead to the increase of fecal water content with(6.62±10.49)% in 4 of 6 rats,but were not statistically significant(P=0.18).The fecal water content decreased with(15.91 ± 13.22)% when treated with lower doses of hydrotalcite in 3 rats,while decreased with 18.31% in only one rat when treated with high doses of hydrotalcite.2.CA could lead to the significant increase of fecal water content with(8.69±8.97)%(P=0.023)in 8 of 9 rats.The fecal water content decreased with(8.07 ± 3.45)%,(7.07 ± 13.12)%,(5.02 ± 6.01)%,respectively,when treated with lower doses,medium doses and high doses of hydrotalcite.3.CDCA could lead to the increase of fecal water content with(6.45 ± 3.49)%significantly(P=0.001)in all 9 rats.The fecal water content decreased with(7.66±6.82)% significantly(P=0.010)in 7 of 9 rats,when treated with medium doses hydrotalcite.4.CDCA mixed with CA could lead to the increase of fecal water content with(9.63±6.11)% significantly(P=0.001)in all 9 rats.The fecal water content decreased with(7.03±7.27)% significantly(P=0.020)in 7 of 9 rats,when treated with medium doses hydrotalcite.5.Bile or bile acid could lead to the increase of concentration of fecal total bile acid with(144.65 ± 129.06)? mmol/L significantly(P=0.003)in 10 of 12 rats.The concentration of fecal total bile acid decreased with(100.57 ± 120.64)? mmol/L significantly(P=0.015)in 7 of 9 rats,when treated with doses hydrotalcite.ConclusionsCA,CDCA and CDCA mixed with CA could lead to the decrease of stool consistence and the increase of fecal water content in SD rats,which could be released on the treatment of hydrotalcite with the capacity to absorb bile acid in lower doses,mediate doses and high doses,but the effects of lower doses,medium doses of hydrotalcite tends to be better.Thus a reference has been provided for the treatment of human bile acid diarrhea.
