Effects Of Jie Du Tong Luo Recipe On Col?, Col ? And MMP-9 In Myocardial Tissue Of MF Rats

Objective:To observe the effect of Jie du Tong luo Recipe on Col I,ColIII(collagen?)and MMP-9(MMP-9)in myocardium of myocardial fibrosis(MF)rats,and to explore the effect of Jie du Tong luo Decoction on myocardial fibrosis The possible mechanism.Methods:30 adult SPF grade SD male rats were randomly divided into normal control group,model group,ACEI group and Jie du Tong luo high,medium and low dose group,each group of 5.The control group was given 0.005g/kg body weight saline,model group,ACEI group and Jie du Tong luo high,medium and low dose group given intraperitoneal injection of 0.005g/kg body weight isoprenaline(isoprenaline,Iso)modeling,starting from the day of modeling,The Jie du Tong luo low,medium and high dose groups were given daily concentration of 1 g/ml of Jie du Tong luo original drug suspension 10,20,30 ml/kg body weight,ACEI group to the concentration of 0.5mg/ml of captopril suspension 10ml/kg body weight,the model group and the normal control group with the same amount of distilled water gavage,were 9 o'clock every day gavage 1,for 14 days.The rats were sacrificed 24 hours after the last time,and the rats were sacrificed.Heart paraffin sections were stained with HE staining and MASSON staining was used to observe the hyperplasia of collagen.The content of hydroxyproline was determined by digestion method.The expression of MMP-9 in myocardium of rats was detected by reverse transcription polymerase chain reaction(RT-PCR).Results:Compared with the control group,the myocardium of the model group showed typical myocardial fibrosis,the Jie du Tong luo recipe group and the ACEI group,the myocardial fiber was not thickened,there was no obvious gap between the muscle bundle,apical and subendocardial Only a small amount of collagen fibers calm.Compared with the model group,the effect of Jie du Tong luo recipe group and ACEI group on the decrease of Hydro content in myocardium was significant(P<0.05).Compared with the model group,the expression of col? and col? in ACEI group and Jie du Tong luo group decreased(P<0.05).Compared with the model group,the expression of MMP-9 mRNA in ACEI group and Jie du Tong luo group was significantly increased(P<0.05).Conclusion:Jie du Tong luo can improve the myocardial function induced by isoproterenol in MF rats by regulating the levels of col?,col? and MMP-9,and its mechanism may be related to the increase of MMP-9 expression.
