The Study Of Jieduyizhifang Influence Amyloid-β Which Aggregated On The Mode Transgenosis C.elegans Of Alzheimer’s Disease

Purpose: There will be two studies which research Jieduyizhifang influences AD C.elegans behavioristics and morphology,and on one side,discussing different concentrations of Jieduyizhifang’s influence on AD C.elegans’ paralysis,then getting relatively better concentration which will protect AD C.elegans mostly,and on other side,discussing inhibiting effect of Aβ on C.elegans’ head in different concentrations of Jieduyizhifang.Method:1,Behavioral experiment:There are four groups which respectively are 25mg/L、50mg/L、100mg/L of Jieduyizhifang and one controll group,each group get 30 C.elegans,they wil be trained 48 h in 16℃,and then move them to 25℃ in order to inducing nervous system injuried.We will observe and record the datas every four hours,this experiment will be repeated three times.2,Morphology experiment:There are four groups which respectively are 25mg/L 、50mg/L 、 100mg/L of Jieduyizhifang and one controll group,each group get 25-30 C.legans,hey wil be trained 48 h in 16℃,and then move them to 25℃,we will take them under fluorescence microscope in order to observing the plaque sedimentation on C.elegans’ head in the third day and the eighth day,and then recording datas,this experiment will be repeated twice.Result:1,The study of behavioristics manifests that 25mg/L、50mg/L、100mg/L groups which are processed with Jieduyizhifang both have significant variation with control group which is processed with nothing(P<0.05),and the rate of paralysis in 25mg/L groupis lowest compared with control group.After analyzing in “Two-way ANOVA ”,the rate of paralyzed in 25mg/L group which is processed with Jieduyizhifang has significant variation from control group at the time of 40 hours,and maintains from 40 hours to 76 hours.2,The study of morphology manifests that there are more sedimentation of Aβplaque in the group of non-processed.While the groups which is processed with Jieduyizhifang has less sedimentation.Conclusion: Jieduyizhifang could realy could improve the paralyzed C.Elegans of AD caused by Aβaggregated and reduce the plaque sedimentation on C.Elegans’ head.
