The incidence of lung cancer and mortality rate was increasing year by year in the global.The clinical treatment of tumor have not specially selective,killing tumor cells either also damage normal cells,the normal human body caused by some side effects,Resulting in poor treatment of mortality.Therefore,An efficient,low toxicity targeted drug delivery system for malignant tumor site is of great significance.At the same time,the study showed that the high expression and activation of epidermal growth factor receptor(EGFR)in a variety of malignant tumors,of which about 80%of non-small cell lung cancer EGFR in the presence of high expression.The non-small cell lung cancer molecular targeting drugs can inhibit EGFR.In this study,we propose a dual-targeted drug delivery system for the targeted therapy of non-small cell lung cancer.The main research contents are as follows:1.We choose the dextran to modified Fe3O4 magnetic particles,and make use of different characterization methods to evaluate the magnetic properties of the magnetic particles,physical and chemical properties.It was confirmed that the magnetic particles of dextran had the characteristics of uniform particle size,large specific surface area and strong magnetic response.The cell viability of A549 cells after different concentrations of dextran magnetic particles in CCK-8 experiment showed that the magnetic particles were a kind of biocompatible,safe,reliable and low toxicity Drug targeting vector.2.Experimental designed the monoclonal antibody(C225)and the small molecule chemotherapeutic drug(Dox)were simultaneously loaded to the dextran magnetic particles to prepare the immunomagnetic composite particles Dox-NPs-C225.The Dox-NPs-C225 was characterized by FITC,TEM,DLS5 UV,SDS-PAGE and other methods.3.Dox-NPs-C225 biological study;CCK-8 experiment shows Dox-NPs-C225 have more lethal for A549 cells,it also can significantly inhibit the expansion of tumor cells;Apoptosis experiments found that the immuneparticles compared with other experimental groups,the apoptosis of Dox-NPs-C225 was observed by TEM.The results showed that A549 cells were mainly treated with receptor-mediated endocytosis,and the Dox-NPs-C225 particles are ingested into the cells.
