Objective:To investigate the metabonomic variation with colorectal cancer patients by Using NMR.Methods: choosing serum sample from 40 patients diagnosed colorectal cancer and 20 patients diagnosed Colorectal precancerous lesions(includes inflammatory bowl disease and Colorectal adenoma)from First Affiliated Hospital of Xinjiang Medical University,healthy controls serum samples 60 cases.The application of nuclear magnetic resonance technology combined with partial least squares analysis of serum levels in three groups of metabolites.Results:14 compounds of Phenylalanine,tyrosine,lactate,?-glucose,choline,leucine,glycine,?-glucose,creatine,glutamine,glutamic acid,acetyl cysteine,valine,proline acid alaninethe in the serum of colorectal cancer patients are significantly decreased than the healthy group,while ?-hydroxybutyrate,acetoacetate increase.The difference was statistically significant(p<0.05).Leucine,choline,?-glucose in the serum of patients with Colorectal precancerous lesions and the colorectal cancer patient are lower than the healthy group.Other than those three compounds isobutyric acid,betaine,citric acid in the serum of Colorectal precancerous lesions are lower than the healty group.K-ras gene mutant colorectal neoplasms have higher levels of ?-glucose,?-glucose and ?-hydroxybutyrate than wild-type K-ras colorectal tumors.(P <0.05)in The case of K-ras gene mutated different anatomical structure CRC acetoacetic acid,acetone,lactic acid higer in the right side of colon than the left side.Conclusion:1.There are several metabonimic variations between patients with colorectal cancer and the healthy group.2.Changes in serum metabolites in patients with Colorectal precancerous lesionsgroup showed a change from the normal to the transition state of the tumor,the variation in some of the metabolimics can predict tumor indirectly.3.K-ras gene expression and the different types of anatomical parts can cause changes in serum metabolites,and changes of those substances have comment with metabolomic research which may have a certain reference.
