Objective:The purpose of this study is to study the relationship between Wnt/beta-catenin signaling pathway related factors as beta-catenin and GSK-3 beta and the occurrence of hepatocellular carcinoma(HCC)in HBV-related HCC issue.Methods:Twenty cases of patients with primary liver cancer,who were all diagnosed with hepatitis B,were enrolled in this study.According to clinical characteristics,such as age,sex,differentiation,TMN stage,lymph node metastasis,HBeAg,HBV,DNA,these statistics were taken down.All the patients were diagnosed as hepatocellular carcinoma in accordance with postoperative pathologic examination.Twenty case of HCC tissues and the paracancerous tissues from the same patient were collected in the operation room.The specimens were labeled and stored in minus eighty Celsius refrigerator for extraction of total RNA and protein.Then,the expression level of mRNA and protein between ?-catenin and GSK-3? were detected in the two groups of HCC tissues and paracancerous tissue,by Quantitative Real-time PCR and Western Blot.The difference of the expression of mRNA and protein in the two groups was analyzed.Results:1.The expression level of mRNA of ?-catenin in HCC tissues was higher than that in Paracancerous tissue,and the difference was statistically significant(P < 0.05).The expression level of of protein of ?-catenin in HCC tissues was higher than that in Paracancerous tissue,and the difference was statistically significant(P < 0.05)2.The expression level of mRNA of GSK-3? in HCC tissues was lower than that in Paracancerous tissue,and the difference was statistically significant(P < 0.05).The expression level of protein of deactivated GSK-3? in HCC tissues was higher than that in Paracancerous tissue,and the difference was statistically significant(P < 0.05).Conclusion:1.The expression level of ?-catenin and deactivated GSK-3? in HCC tissues was up-regulated,which provide certain experimental basis for the molecular targeted therapy of liver cancer.
