| Objection:To investigate the effect of apolipoprotein M(ApoM)on the incidence and development of deep vein thrombosis(DVT)in mice.Methods:With base quenched probe techniques,we confirmed the genotypes of mice as ApoM wild type(ApoM+/+)or ApoM knockout type(ApoM-/-).24 mice(12 for each genotype)were divided into two groups.The inferior vena cave was ligated below the renal veins in the DVT group,and the other group is sham group.Inferior vena cave in mice was ligated for 6 hours and 48 hours,then the thrombus’s weight and length was measured in the DVT group,and the plasma coagulation indexs(PT,APTT,FIB,TT)in the sham and DVT groups.The expression of TF,TNF-α,IL-1βand IL-6 in inferior vena cave were detected by RT-PCR in the sham and DVT groups.Results:The incidence of thrombus in ApoM+/+mice and the ApoM-/-mice didn’t have significant difference in the DVT group(P>0.05).The weight and length of thrombus in ApoM-/-mice were significantly higher than those in ApoM+/+mice(P<0.05).Nevertheless,the activated partial thromboplastin time(APTT)of ApoM+/+mice was significantly lower than ApoM-/-mice(P<0.05).Conclusion:ApoM may inhibit the development of DVT by prolonging the APTT of mice.But it has no dominant effect on the incidence of DVT. |
