The Effects Of Annexin?/S100A9/vimentin Interactions In The Invasion And Metastasis Of NPC

Objective:Experiments were used to confirm whether NPC cells were able to form Annexin I / S100A9 / Vimentin protein interactions;the expression of Annexin I / S100A9 / Vimentin in NPC tissues and cells was detected by immunohistochemistry and western blot,and then analyze the expression of Annexin I/S100A9/Vimentin in NPC tissues and cells.The study was performed to establish an up-regulation and down-regulation of Annexin I expression in NPC cells and analyzing the effect of up-regulation or down-regulation of Annexin I on the expression of S100A9 / Vimentin in NPC tissues,cells and analyzing the effect in the invasion and metastasis of NPC.Our experiments will initially elucidated the role of Annexin I in regulating the invasion and metastasis of NPC through S100A9/Vimentin,and provide a clues for the research of the prevention and cure of NPC.Methods: in this research,the interaction between Annexin I /Vimentin / S100A9 protein in nasopharyngeal carcinoma 6-10 B cells was confirmed by immunoprecipitation combined with Western blot;the tissue specimens of NPC have been collected to detected the expression of Annexin I / S100A9 / Vimentin in nasopharyngeal carcinoma by immunohistochemical staining;NPC cell lines 5-8F with higher transfer,6-10 B with lower transfer,CNE1 with Well differentiated and CNE2 with poorly differentiated were selected,and the expression of Annexin I / S100A9 / Vimentin was analyzed by Western blot in abovefour kinds of NPC cell lines.Then recombinant plasmids Annexin ?(+)and Annexin-?-RNAi(+)were constructed,and transfected into 5-8F and6-10 B cells by Lipofectamine? 2000 respectively.Finally,Western blot,transwell and Wound healing assay were used to detect the effect of Annexin ? up-regulation or down-regulation on the expression of S100A9 / Vimentin and the biological behavior in recombinant NPC cell lines.Results: 1.The interaction between Annexin I / Vimentin / S100A9 protein in nasopharyngeal carcinoma 6-10 B cells was confirmed by immunoprecipitation and Western blot,the results showed that there were interaction between Annexin I,S100A9 and Vimentin in NPC cells.2.In NPC tissues,S100A9 was negatively correlated with Annexin I expression(r=-0.640,P< 0.01),Vimentin was negatively correlated with Annexin I expression(r=-0.422,P< 0.05),While S100A9 was positively correlated with Vimentin expression(r=-0.390,P< 0.05);3.The expression of Annexin I in CNE1 and 6-10 B cells was stronger than the expression of Vimentin and S100A9,while the expression of the three proteins in CNE2 and 5-8F cells was opposite;4.Annexin ?down-regulation could may increase the expression of Vimentin and S100A9,and enchance the in vitro invasive ability in the NPC cells by Vimentin / S100A9 interaction.Conclusions:(1)In NPC cells,Annexin I / Vimentin / S100A9 can form protein interaction;?Annexin ? may inhibit the invasion and metastasis of NPC cells through the interaction with S100A9 / Vimentin protein...
