Objective:1.To observe the effects of resveratrol on blood sugar after OGTT and the expression of 3-phosphoinositide dependent protein kinase-1(PDK-1).2.To observe the effects of resveratrol on the expression of 3-phosphoinositide dependent protein kinase-1 phosphorylated protein kinase B and Akt in the MIN6 cell lines.3.To observe the effects of resveratrol on the ratio of proliferation.4.To observe the effects of resveratrol on the apoptosis.5.To observe the effects of resveratrol on the level insulin in extracellular fluid.Methods:1.the blood was collected from the orbital vein after OGTT and was measured by biochemical detector,the expression of 3-phosphoinositide dependent protein kinase-1(PDK-1)were measured in the three groups after 4 weeks intervening.2.The expression of PDK-1,p-Akt and Akt was measured by Western blotting in the four groups.3.The cell proliferations were detected by CCK-8 kit.4.The cells apoptosis were detected by cytometry.5.The levels of insulin in cell supernatant were detected by ELISA.Results:1.Compared with diabetic group,the OGTT blood glucose area under the curve decreased significantly in db/db mice intervented by resveratrol(P<0.05),the expression of PDK,1 protein increased[positive rate(1.74 ± 0.47%)vs(0.57±0.32%),P<0.01].2.Compared with HG group,the expression of PDK-1,p-Akt protein increased in HG+Res group,Compared with the HG+Res group,the expression of PDK-1 and p-Akt protein decreased in HG+Res+Wo group,the difference were statistically significant,P<0.05.3.Compared with HG group,the cell proliferation[(1.02 ± 0.09)vs(0.71 ± 0.10)]increased in HG+Res group.Compared with the HG+Res group,the cell proliferation were decreased in HG+Res+WO group,the difference were statistically significant,P<0.05.4.Compared with HG group,the apoptosis rate decreased[(11.2±1.4%)vs(19.3±2.1%)](P<0.01)in HG+Res group,Compared with the HG+Res group,the cell apoptosis rate increased in HG+Res+Wo group,the difference were statistically significant,P<0.05.5.Compared with HG group,the insulin levels in cell supernatant[(181.19±30.77)vs(124.95±5.37)pmol/L]increased in HG+Res group,Compared with the HG+Res group,the insulin level were decreased in HG+Res+Wo group,the difference were statistically significant,P<0.05.Conclusion:Resveratrol could increase the proliferation and decrease the apoptosis of in in MIN6 cells.and may play an important role in the protection of ? cells.The potential mechanism of this protective effect is associated with the activation of the PDK-1/Akt signal pathway.
