Laryngeal cancer is one of the most common malignant tumors in China,which seriously threatens people’s health and life.The research progress of traditional Chinese medicine provides the possibility of neoadjuvant chemotherapy,Curcumin is a kind of phenolic pigment extracted from the rhizome of Curcuma plants,which has no obvious side effects on normal cells,but it can improve the sensitivity of tumor cells to chemotherapeutic drugs.In this study,in order to provide experimental evidence for improving the clinical efficacy and reducing the side effects of docetaxel,curcumin and docetaxel were used to treat human laryngeal carcinoma Hep-2 cells to observe the anti-tumor effect of curcumin on chemotherapy.Research methods:The Hep-2 cells were cultured in vitro for pre experiment,curcumin was established with 0mol/L,5 mol/L,20 mol/L,40 mol/L,and 80 mol/L of five groups and taxotere was established with0ng/ml、1ng/ml、25ng/ml、50ng/ml、100ng/ml of five groups,observed the proliferation of Hep-2 cells after 0,24,48 and 72 hours of culture and then select the best concentration of the experiment by MTT experiment.The experiment was divided into four groups: blank control group(only culture solution),curcumin group(20 mol/L curcumin),docetaxel group(25ng/ml docetaxel),mixed group(20 mol/L curcumin + 25ng/ml docetaxel).MTT method was used to detect the growth and proliferation of Hep-2 cells;The morphological changes of Hep-2 cells were observed by inverted microscope.Annexin-V/PI experiment was used to detect the apoptosis of Hep-2 cells.The expression of P-gp in Hep-2 cells was detected by Western-blot.Result:The results of MTT showed that curcumin and docetaxel could inhibit the growth and proliferation of Hep-2 cells,and the mixed group had synergistic effect;Inverted microscope showed that the number of deaded cells of Hep-2 cells in the combined group of curcumin and docetaxel increased,but there was no significant difference between the groups.Annexin-V/PI experiment showed that the apoptosis rate of Hep-2 cells combined with curcumin and docetaxel was higher than that of other groups.Western-blot experiment showed that the expression level of P-gp in Hep-2 cells treated with curcumin and docetaxel was lower than that in other groups.Conclution: Curcumin and Docetaxel both can inhibited the growth and proliferation of Hep-2 cells and induced apoptosis;Curcumin increased the sensitivity of Hep-2 cells to docetaxel and enhanced the killing effect of docetaxel on Hep-2 cells.
