Objective: In this study,we established an animal model of steroid induced osteonecrosis of the femoral head in New Zealand rabbits.Through each animal bone marrow PPAR gamma,osteocalcin m RNA and trabecular bone area ratio,the ratio of empty lacuna,average diameter index of fat cells compared to verify the conclusion of the experiment.To study the mechanism of action of psoralen on steroid induced avascular necrosis of femoral head.Methods: The New Zealand white rabbits were used as the research object,15 male and female,a total of 30.The animal model of steroid induced avascular necrosis of the femoral head was prepared by modified horse serum plus methylprednisolone.The experimental group was divided into blank group,model group and experimental group,each group had 8 animals.The successful packet,began to administration,the blank group and the model group animal with saline gavage,every day in each animal6 ml kg-1 as the standard,the experimental group with the same dose given animal psoralen extract gavage,a total of 8 weeks.Based on OSTEm RNA,PPAR y m RNA and trabecular bone area ratio,the ratio of empty lacuna and fat cell diameter index observation,clear Chinese herbal medicine for the positive effect of rabbit steroid induced avascular necrosis of femoral head.Results: Determination of OSTEm RNA,PPAR y m RNA: compared with the blank group,model group,PPAR gamma m RNA relative expression increased,osteocalcin expression decreased significantly;compared with the model group,the experimental group of PPAR gamma m RNA expression decreased,osteocalcin expression increased significantly.Determination of fat cells: comparison of the average diameter of the blank group and model group the average diameter of fat cells,fat cells in model group the average diameter increased significantly,the difference was statistically significant(P<0.05);compared with the control group and the experimental group,the average diameter of fat cells did not change significantly,the difference was not statistically significant(P>0.05);compared with the model group the experimental group,the difference was statistically significant(P<0.05).Compared with the control group,experimental group,model group,trabecular bone area was significantly reduced,the difference was statistically significant(P<0.05);trabecular bone area in the experimental group and the blank group did not change significantly,no statistically significant difference between the groups and the actual(P<0.05).The measurement of trabecular bone area: the difference of trabecular area between the groups was compared with single factor test,the difference was statistically significant(P < 0.05).In the model group and blank control group,experimental group in trabecular bone area,at a low level,significant differences were statistically significant(P<0.05);and the trabecular bone area of experimental group and control group were at a high level,virtually no statistically significant differences between the two groups(P>0.05).Determination of the empty bone lacuna rate: univariate variance test,compared the rate of empty lacunae between groups,the difference was statistically significant(P < 0.05).In the model group compared with the control group and the experimental group,the rate of empty lacunae at the high level,the differences were statistically significant(P<0.05);and the empty lacuna in the experimental group and control group were at a low level,virtually no statistically significant differences between the two groups(P>0.05).Conclusions: A rabbit model of steroid induced necrosis of the femoral head by horse serum plus methylprednisolone preparation;the expression of OSTEm RNA and PPAR gamma m RNA by psoralen has remarkable effect,inhibition of bone marrow stromal stem cells to differentiate into adipocytes;improve the steroid induced femoral head Buguzhi lacuna than negative influence rate,fat cell diameter and bone trabecular area ratio.
