Objective:Cyclocarya paliurus contains a variety of active ingredients that can regulate the body's physiological functions.In this paper,the water-soluble extract of C.paliurus(Batal.)Iljinskaja was used as the research object.The bioactivity and hypoglycemic mechanism were studied from two levels of animals and cells and the chemical constituents of the fingerprints were analyzed by HPLC in order to explore the mechanism of hypoglycemic effect of water soluble extract of C.paliurus.Methods: Water soluble components of C.paliurus were obtained by using80 ? distilled water soaked the leaves of C.paliurus.The chemical constituents of the water-soluble extract of C.paliurus were analyzed by HPLC.After a week for adapting the environment,50 ICR mice were randomly and equally divided into the following 5 groups: blank group,streptozotocin(STZ)group,STZ + fatty diet group,STZ+QQL group,and STZ +fatty diet +QQL group.Mice in blank group,STZ group and STZ+QQL group were fed with normal diet;mice in STZ + fatty diet group and STZ +fatty diet +QQL group were fed with fatty diet.Animals in STZ group,STZ plus fatty diet group,STZ+QQL group and STZ +fatty diet +QQL group were administrated daily with STZ in a dosage of 50 mg STZ·kg-1body weight by intraperitoneal injection for three consecutive days.The experiment was prolonged for67 d.During the experiment fasting serum glucose concentrations of mice in each group were measured by glucose detection kit at day 0,day 7 and day 37 after STZ injection,and fasting serum insulin concentrations of mice in each group were measured by insulin detection kit at day 37 after STZ injection.After the set on of diabetes model,the drinking water of STZ+QQL group and STZ +fatty diet +QQL group were switched into Paliurus aqueous extract.we investigated the effects of aqueous extract of C.paliurus on ?-glucosidase and cell uptaked in vitro by physical and chemical experiments and cell experiment.The results showed as follows:? In order to determine the active ingredients,HPLC chromatographic conditions of the aqueous extract of C.paliurus were explored and its HPLC fingerprint map was obtained.Three of the components were identified as cytidine nucleoside,uridine and guanosine.? In animal model,the aqueous extract of C.paliurus has significant hypoglycemic effect on STZ diabetic model,but has certain hypoglycemic effect on STZ and high fat combined diabetes model and a prolonged effect is not obvious.? In vitro experiments showed that the aqueous extract of C.paliurus can significantly have an effect on the release of reducing sugars by cellulase and a certain inhibitory effect on ?-glucosidase,but it may be not the main reason for reducing the blood glucose level of ICR diabetic mouse.? In the cell model,the aqueous extract of C.paliurus can significantly promote 3T3 cells to uptake.It can be concluded that QQL promotes blood sugar absorption from the cells in mice to reduce the concentration of blood glucose in ICR diabetic mice.
