Objective: To optimize the extraction process of polysaccharides from Cyclocarya paliurus(Batal.)lljinskaja.To observe the effect of Cyclocarya paliurus(Batal.)lljinskaja and its prescriptions on the model of T2 DM and its mechanism of action.Methods: 1.The optimum extraction process of polysaccharides from Cyclocarya Paliurus(Batal.)lljinskaja was studied by single factor experiment and orthogonal test on extraction temperature,solid-liquid ratio and extraction time.2.High-fat diet combined with low-dose streptozotocin induced type 2 diabetes model to detect the basic state,body weight,blood sugar,blood lipids and pathological state of pancreatic tissue.3.Real-time quantitative PCR was used to detect the expression of IRS-1,PI3 K,AKT and GLUT-4 mRNA in the muscle tissue of the quadriceps muscle.Results: 1.Optimization of extraction process of polysaccharides from Cyclocarya Paliurus(Batal.)lljinskaja.The optimal extraction process is that the extraction temperature is 90 ° C,the ratio of material to liquid is 1:40,and the extraction time is 1h.Under this condition,the extraction rate of Cyclocarya Paliurus(Batal.)lljinskaja polysaccharide is the highest,which is 2.45%.2.(1)Basic state of the rats: The rats in the normal control group continued to maintain a stable and good state during the experiment,with sufficient energy,sensitive activity,gentle personality and bright white hair.T2 DM model rats before administration,lazy and less active,violent behavior,hair color yellowing off,obvious eating more,more drink,polyuria,weight loss symptoms.After administration,the symptoms of "three more and one less" in the administration group and the positive control group were alleviated to varying degrees.(2)Changes in body weight of rats in each group: There was no significant difference in body weight between the groups before modeling.After 4 weeks of feeding high-fat diet,the body weight of each group increased,especially in the model group.After injection of streptozotocin(STZ),the weight of the other groups was significantly reduced compared with the normal control group.After 8 weeks of administration,the body weight of the administration group(except for the high dose group of Cyclocarya paliurus(Batal.)lljinskaja polysaccharide),the positive control group,and the model group increased.(3)Changes in blood glucose of rats in each group: There was no significant difference in blood glucose between the groups before injection of STZ.Before administration,the blood glucose of the model rats was significantly increased.After administration,compared with the model group,the low dose group of Cyclocarya paliurus(Batal.)lljinskaja polysaccharide,the low,medium,and high dose groups of Cyclocarya paliurus(Batal.)lljinskaja prescriptions and the positive control group had significant blood glucose decline.The blood glucose of the rats in the high dose group of Cyclocarya paliurus(Batal.)lljinskaja polysaccharide and Cyclocarya paliurus(Batal.)lljinskaja water extract group decreased slightly.(4)Changes in blood lipids in rats in each group: Compared with the normal control group,rats in the model group had elevated total cholesterol(TC),triglyceride(TG),low-density lipoprotein cholesterol(LDL),and high-density lipoprotein cholesterol.(HDL)fell.Compared with the model group,the TC,TG and LDL of the drug-administered group and the positive control group were smaller than the model group,and the HDL was larger than the model group.(5)Morphological observation of rat islet cells: In the normal control group,the islets in the pancreas were round or elliptical,with a large number of cells,large and round nuclei,neatly arranged,and clear boundaries.In the model group,the number of islet cells was significantly reduced,the islet structure was severely damaged,the shape was irregular,and the boundary was blurred.The number of islet cells was significantly reduced,arranged disorderly,with atrophy,fibrosis and inflammatory infiltration.The islet structure of the drug-administered group and the positive control group was relatively intact,the boundary was clear,and it was nearly elliptical.The islet cells were arranged neatly,the cell morphology was complete,the cytoplasm was abundant and the nucleus was in the middle,and only a few cells were degenerated or necrotic.3.Compared with the normal control group,the expression levels of IRS-1,PI3 K,AKT and GLUT-4 in the model group were down-regulated.Compared with the model group,the expression levels of IRS-1,PI3 K,AKT and GLUT-4 significantly up-regulated of the positive drug group,the low dose group of Cyclocarya paliurus(Batal.)lljinskaja polysaccharide and the Cyclocarya paliurus(Batal.)lljinskaja prescriptions.Conclusion: 1.The optimal extraction process of polysaccharides from Cyclocarya Paliurus(Batal.)lljinskaja was extraction temperature of 90 ° C,ratio of material to liquid of 1:40,and extraction time of 1 h.2.Cyclocarya Paliurus(Batal.)lljinskaja and its prescription can have hypoglycemic effect on T2 DM rats,and also have a regulating effect on blood lipids.3.Cyclocarya Paliurus(Batal.)lljinskaja and its prescriptions can improve peripheral insulin resistance.The mechanism may up-regulate the expression of IRS-1,PI3 K,AKT and GLUT-4 mRNA,activate peripheral islet resistance and enhance islet function by activating PI3K/AKT signaling pathway..
