Objective:This experiment through the radiation group,scald group,gross observation of the wound was blank group three groups in the process of wound healing,observation of fibroblast distribution in wound tissue pathology,the number of changes,immunohistochemical detection of vimentin expression(fibroblast cytoskeletal protein)and other aspects of the differences,changes of fibroblasts in wound healing to explore the acute wound beta ray skin II degree injury healing.Methods: 102 SD rats provided by the university of South China,weighing 200 ± 5.45 g,single cage for one week,were randomly divided into three groups.Radiation group: n = 48,6MeV high-energy electron beam linear accelerator single exposure to the hips bare skin,the total receiving dose of about 35 Gy,irradiation diameter of 2cm,the establishment of acute skin ?-ray deep ?° injury wound model;Scald group: n = 48,the choice of diameter 2cm test tube set,the test tube filled with hot water,placed in the 90 ?electric thermostatic water bath,so that the bare hips and test tube water contact,to maintain 9s,the establishment of skin Deep ?° thermal scald animal model;Blank group: n = 6,without any processing.At the time points(after the establishment of the model 1,4,7,14,21,28,35 days)to take the rat hips wound area or a considerable area of skin tissue,the three groups of tissue to be taken to perform histopathological sections,HE staining and immunohistochemistry were used to observe the changes of fibroblasts in dermal tissues at different time points(the wounds in the experimental study healed naturally without external intervention).Results:1,general observation: rats after irradiation,wound erythema,swelling,and disposition became irascible,hnecrotic skin off 14-21 days,21 days,the wound began to shrink gradually and new skin had grew,28-35 days,the wound healed gradually.In comparison,Scald group in the burn after the skin white,swelling,crusting,14 days when the wound skin crusted,and began to fall off,21 days when the newborn skin produced,the wound basically healed on the 28 day.2 HE staining of wound tissue:after 4 days of treatment,scald group,the fibroblasts of scald group had proliferation and migration,while irradiation group had no change;in the scald group,fibroblasts,7-14 increased rapidly,migrated,and peaked during this period,then decreased gradually and transformed into mature fibroblasts;but during the 14-35 days after radiation,the fibroblasts increased slowly and the peak was not seen in the irradiated group;the pathological in the blank group were not changed.3?Immunohistochemical results:the IOD of vimentin increased in seventh days after irradiation,but the progress of increase was slow and smooth,and there was no obvious peak,trough and so on;after empyrosis,the IOD value of fibroblasts in scald group began to increase and increased rapidly during the days of 4-7 and peaked in the period of 7-14 days,then decreased gradually after 14 days;there was no changes for cells in the blank.Conclusion:The wound of acute ?-ray skin second degree injury heal slower than deep II degree scalded wound of skin.Radiation damage in fibroblasts differentiation,migration ability than scalded fibroblastsis lower and can not form a clear peak;2?the experiment has approve that fibroblasts paticipated in wound healing.
