Objective To study the effect of recombinant human acidic fibroblast growth factor (rhaFGF) and recombined human epidermal growth factor (rhEGF) on wound healing of rabbits'ears with full-thickness skin injury.Methods①Models of rabbits'ears with full-thickness skin injury were made and divided into groups according to concentration gradient experiment: group A(rhaFGF 60U/cm~2), group B(rhaFGF 90U/cm~2), group C (rhEGF 20U/cm~2), group D (rhEGF 40U/cm~2)and group E (equivalent normal saline). According to the result of the concentration gradient experiment to set the concentration of the combination experimental. The models of combination experimental are same as the former. Drug combination models were randomly divided into four groups: group F (rhaFGF 60U/cm~2+rhEGF 20U/cm~2), group G (rhaFGF 60U/cm~2), group H (hbFGF 60U/cm~2)and group I (equivalent normal saline).②According to different groups,the wounds were given the drugs immediately after the surgery. Then,these reagents were given every two days.③Experimental evaluation. Wound healing rate and time were respectively recorded at the 3rd, 7th, 10th, and 14th day by hyaline membrane tracing. Optimum reagent concentration was determined by making comparison of wound healing rate and time in different groups and drug combination experiment was designed according to the result. Models of drug combination were made in the same way as that in single drug groups.In order to observe the wound healing, the wound tissue of drug combination which was detected the expression of vascular endothelial growth factor (VEGF) and matrix metalloproteinases- 1 (MMP-1) by immunohistochemistry and stained by hematoxylin and eosin was taken in the above time points. And the tissue also was detected the expression of VEGF mRNA by everse transcription polymerase chain reaction at the same time.Results①In concentration gradient experiment, wound healing time in group A was 13.57±0.40 days, which has statistical significance compared to group E( P <0.05). Wound healing rate in group A was also statistically different from that in group E( P <0.05) at the 7th, 10th and 14th day. In drug combination experiment, wound healing time in group F was 13.33±0.12 days, which had statistical difference compared to that in group I( P <0.05). Wound healing rate in group F was also statistically different from that in group I( P< 0.05) at each time point.②HE staining showed that at the 3rd and 7th day after modeling, growth of fibroblasts and capillary was more vibrate in group F and H than that in group G and I. At the 14th day after modeling, wound was almost healed in group F and fibroblasts and capillary were relatively rare compared to other groups. Histological status in group G was between that in group F and group H. All other groups were superior to group I in terms of the wound healing state.③Every group begun to express MMP - 1 at the 3rd day after modeling, which has no obvious difference with statistical difference compared to that in each other. At the 7th day, MMP-1 of every group reached a peak value. Group F had obvious statistical difference compared to group I (P <0.01).Group G and H had obvious statistical difference compared to group I ( P< 0.05).At the 14th day, the expression of MMP-1 reduced in every group. Group F and H had statistically lower than that in group I (P< 0.05). There was no statistical difference between group I and H (P>0.05), though group H had the highest expression among all the group.④The expression of VEGF in all groups had no statistical difference at the 3rd day(P>0.05). It reached a peak value at the 7th day. Group F had obvious statistical difference compared to group I (P <0.01).Group G and H had obvious statistical difference compared to group I ( P <0.05).VEGF of group F had statistically lower than that in group I at the 14th day ( P <0.05) .There was no statistical difference between group I and H (P>0.05), though group H had the highest expression among all the group.⑤The expression of VEGF mRNA in all groups had no statistical difference at the 3rd day(P>0.05). The combination can stimulate the growth of granulation tissue, capillary formation and re-epithelialization. In the later stage of wound healing, capillary and fibroblasts grow slower than in other groups, which may reduce the possibility of scar formation.②The possible mechanism of group F promote wound healing may be raised the expression of VEGF mRNA, VEGF, MMP-1 in early. The possible mechanismin of group F reduced scar formation may cut down the value of VEGF mRNA,VEGF,MMP-1 in late.③D rug combination is better than the singnal in wound healing.
