Aim: To investigate the effect and potential mechanism of magnesium isoglycyrrhizinate on Con A-inducedacute liver injury in mice.Methods: Con A(20mg/kg)was injected through tail vein on Bal/C mice to induce acute liver injury.And different concentrations of MgIG was injected to protect in advance.All serum and liver tissues in different group were got at 2h,8h and 24 h after Con A injection.The levels of liver enzymes(ALT and AST)and cytokines(IL-6 and TNF-?)were detected in serum.The level of liver enzymes and pathological change were observed and compared in different groups.through methods of Elisa,Real time PCR,Western blot and immunohistochemistry,the expression of TRAF6 and JNK signal pathway related genes,as well as apoptosis and autophagy activity in different groups were detected to explore the potential mechanisms.Results:MgIG could decrease the level of ALT and AST and ameliorate the pathological change in acute liver injury;reduce the expression ofinflammation factor(IL-6,TNF-?)in serum and liver tissue;Reduce the expression of TRAF6 and inhibit the activation of JNK signal pathway;Reduce the expression of Baxand Beclin 1,and increase the expression of Bcl – 2.Conclusion:MGIG may inhibit TNFR6 and JNK pathway,lower levels of intrinsic apoptosis and autophagy to ameliorate Con A-induced acute liver injury.
