Objective:The present study aims to evaluate the hepatoprotective effect and possible mechanism of the Magnesium Isoglycyrrhizinate(MgIg) in acute liver injury by measuring the protein expression level of ERK/MAPK signal pathway as well as its phosphorylation level.Methods:1.We Collected 31 cases patients with drug-induced liver injury and measured the variations of Alanine Amino Transaminase (ALT), Aspartate Amino Transaminase (AST), Interleukin-6 (IL-6) and Procalcitonin (PCT) before and after they accepted the treation with magnesiumiso glycyrrhizinate by extracting venous blood samples.2.The acute liver injury model of wistar mouce were established by lavageing with 15%CC14 peanut oil solution. The models were regarded as experiment groups and randomly divided into three groups including liver injury model group, Magnesium Isoglycyrrhizinate treatment group and normal saline treatment group. While the same batch number Wistar mouce were selected as normal control group. Two weeks later, all groups mouce were executed, then liver tissues were taken out and made into liver Tissue Micriarray(TMA), which were stained with hematoxylin and eosin (H&E) and examined the differences of the liver tissues pathology among these groups by a light microscope. Then the expression levels of ERK and p-ERK in each group of the liver tissues were investigated by immunohitochemistry technology and analyzed the pathologic pictures by Microscopic Image Analysis System. Moreover, venous bloods from all of the mouce were collected to measure the levels of ALT and AST by a Biochemistry Automated Analyzer, and interleukin-6 (IL-6) with Roche Immunoassay Test Kit. According to the levels of ALT、AST and IL-6, we estimated the hepatitis degree of the each group.Results:1.Differences of ALT, AST, IL-6 and PCT in 30 patients with drug-induced hepatitis before and after treated with magnesium isoglycyrrhizinate were statistically significant (P<0.05). 2.Compared with the MgIG group and control group, the model group of the mean gray value(OA) and percentage of positive mouce cells of ERK and p-ERK expression were significantly increased. While compared with the NS group, MgIG group of the OA and percentage of positive cells was remarkable decreased. Both there are difference by the statistically analyzed (P<0.05). The significantly increased levels of IL-6, ALT, AST in model and NS groups compared with the outstanding declined MgIG group, there was statistically significant difference (P<0.05).Conclusion:1.Magnesium isoglycyr-rhizinate can significantly improves liver function in patients with drug-induced liver inflammation. While IL-6 and PCT can serve as effective serological markers to determine efficacy.2.Magnesium Isoglycyrrhizinate might alleviate liver injury by inhibition of ERK signal pathway.
