Objective:We study the effect of estrogen on the expression of TNF-?and IL-1?in LPS-induced inflammation model of mouse endometrial stromal cells,and investigate whether estrogen has antiinflammatory effects in the mouse endometrial stromal cell inflammation model.The aim is to demonstrate the anti-inflammatory effects of estrogen,So as to provide experimental basis for the clinical application of estrogen in the treatment of endometritis.Methods:We choose the Chinese Kunming mice of Female,They are6-8 weeks old without produced and weight 25-30g.The rat endometrial stromal cells were isolated and purified by enzymatic digestion,filtration.We culture the cells in vitro.Cell phenotype was identified by immunocytochemistry.And we establish the LPS induced mouse endometrial stromal cell inflammation model.On this basis,we used different concentrations of estrogen for different time to carry out anti-injury study,while observing the cell morphology under the microscope.The levels of TNF-?and IL-1?in each group were detected by RT-qPCR method,analyzing the differences in each group with statistics,and then determine the optimal concentration of estrogen and the optimal role of time.Simultaneously analyzed the effects of estrogen on the expression of TNF-?and IL-1?in LPS-induced mouse endometrial stromal cells.Results:The mouse endometrial stromal cells were successfully isolated and identified,The best concentration of LPS-induced mouse ESCs Inflammation was 100ng/ml.The results of real-time fluorescence quantitative PCR showed that the expression levels of TNF-?and IL-1?mRNA were significantly different in 100ng/ml LPS group compared with the control group at different time?p<0.01?.There was no significant difference between estrogen and control group at different time?p>0.05?,There was no significant difference between E210-9+LPS and LPS group at different time?p>0.05?,and The expression of TNF-?and IL-1?of E210-8+LPS was significantly different from that of LPS group?p<0.01?.The expression of TNF-?and IL-1?in E210-7+LPS,E210-6+LPS group was significantly lower than that in LPS group,the difference was statistically significant?p<0.01?.Conclusion:Estrogen can downregulate the expression levels of inflammatory factors TNF-?and IL-1?in a concentration-dependent manner,indicating that estrogen has anti-inflammatory effect.
