Objective: Through the establishment of vascular dementia(VD)rat model,observe the VD rats' behavioral changes after the intervention of Fuzhi capsule,and the changes in protein expression level of drebrin and mammalian target of rapamycin(m TOR)in the hippocampus of rats in each group,study the mechanism of Fuzhi capsule improving VD.Methods:Select 36 male SD rats in SPF level weighing about(300±20)g,and screen all the rats in the Morris water maze after the adaptive feeding,to remove the rats cannot complete behavioral test.The rats with comepleted behavioral test are grouped into blank control group and model groups by random number table method in Excel,and the VD model was prepared by permanent ligation of bilateral common carotid artery ligation(2-VO)method from the rats in model groups.The model groups that survived are screened in the Morris water maze again,the successful VD model is the rats with obvious learning and memory disorder,and divide them into model control group ?Aricept group and Fuzhi capsule high?medium?lower-dose group by random number table method in Excel.After the drug intervention in 30 day,conduct the place navigation test in each group by the Morris water maze,and the results are accorded to evaluate the learning and memory abilities of rats in each group.Cut off the head to take out the brain after the behavioral test,and use Western blotting method to detect the protein expression level of drebrin and m TOR in the hippocampus of rats.Results:1.Result of the Morris water maze:The strategy of space search of rats in blank control group and drug intervention group are mostly linear,but the rats in model group are mostly marginal or random.In place navigation test,model control group have significantly longer escape latency as compared with the blank control group(P<0.01).After the drug intervention treatment of Fuzhi capsule,the escape latencies of high ?medium?lower-dose groups are all shorter than the model control group,the differences are statistically significant(P < 0.05);there are no significant difference among the escape latencies between high-dose group and Aricept group(P>0.05).2.Result of the Western blotting:2.1 Compared with the blank control group,the expressions of protein drebrin in model control group are significantly reduced(P<0.01).After the drug intervention treatment of Fuzhi capsule,the expressions of protein drebrin in high?medium?lower-dose groups are higher than the model control group in different degree,the differences are statistically significant(P < 0.05);there are no significant difference among the expressions of protein drebrin between high-dose group and Aricept group(P>0.05).2.2 Compared with the control group,the expressions of protein m TOR in model control group are significantly increased(P<0.01).After the drug intervention treatment of Fuzhi capsule,the expressions of protein m TOR in high?medium?lower-dose groups are lower than the model control group in different degree,the differences are statistically significant(P < 0.05);there are no significant difference among the expressions of protein m TOR between high-dose group and Aricept group.(P>0.05).Conclusion: 1.Through the 2-VO method to prepare the rats model of VD caused by cerebral chronic hypoxia-ischemia,after the drug intervention treatment of Fuzhi capsule,VD rats' cognitive impairment are effectively improved.2.Chronic hypoxia-ischemia due to the injury of VD rats' hippocampal neurons,the drebrin protein levels are decreased,after the drug intervention treatment of Fuzhi capsule,the neurons damage are reduced,and the learning and memory abilities are improved,the mechanism might be through the Fuzhi capsule regulate the body's m TOR signaling pathways and control the expression of m TOR.
