In this paper,we give a brief overview about the recent applications of SERS technology on the detection of little mount of biochemistry molecular.In current,mast cell degranulation detection technology is time-consuming,short half-life,poor color effect.To overcome the disadvantages of the existing techniques,we employed a new method which is based on SERS technology combined with Principal Component Analysis-Linear Discriminant Analysis(PCA-LDA).This method may be simply,repidly,undestructively to analyse degranulation of mast cell.To date,the mechanism of laser acupuncture is unclear at present,which have hampered its development.In this thesis,we try to learn this mechanism start from the relationship between laser and mast cell degranulation.Different irradiation time and different laser wavelength were exposed on mast cell,and the corresponding cultured supertant were collected for SERS measurement.The main research results are follows:1.SERS technology were used to detect the cultured supertant of mast cell(P815)degranulation in different degree.PCA-LDA were applied to the obtained SERS spectroscopy.Each group seperate well from the nonlaser group.The result show that SERS combined with PCA-LDA has more potential for detection of mast cell degranulation in different degree.2.SERS technology were applied to the cultured supertant of mast cell(P815)degranulation in different irradiation time.The release rate of ?-glucosidase were measured.The result showed that mast cell degranulation in different degree for different irradiation time.3.SERS technology were applied to the cultured supertant of mast cell(HMC-1)degranulation in different Laser wavelength.The analyis of PCA-LDA can distinguish the spectral data well with high sensitivity and specificity.The result showed that different wavelength laser may interact with mast cell in different.
