Along with the improvement of animal husbandry and people’s living standards,In animal husbandry the use of pesticides and veterinary drugs is getting higher and higher.Tetracycline antibiotics due to its antimicrobial spectrum were widely used to feed additives of the treatment of animal diseases and promote animal growth.However,when the unreasonable use of tetracycline antibiotics cause residues in animal-derived foods,which lead to exposure to the population,endangering human health,which is the world attention.To explore tetracycline antibiotics in foods of animal origin and its residual exposure assessment,we conducted the following studies.Part I: Methodological Establishment of Tetracycline Antibiotics in Animal origin FoodObjective: To establish a highly efficient sample pretreatment,extraction,purification and instrumentation system for the simultaneous detection of tetracycline antibiotics in animal origin foods by high performance liquid chromatography-mass spectrometry(LC-MS / MS).Methods: The tetracycline antibiotics in animal food were extracted by EDTA buffer solution by solid phase extraction technique and purified by Bond Elut Plexa column,were detected by LC-MS/MS.Results: Four kinds of tetracycline antibiotics had a good linear relationship,with sharp peak shape,no impurity peaks,and r > 0.99.The detection limit of chlortetracycline,oxytetracycline,doxycycline and tetracycline was 0.1 μg / kg.Meat,milk,eggs and aquatic products of the target tetracycline plus standard recovery were in the 75%-130%;instrument reproducibility RSD <1%.Conclusion: The method has the advantages of high selectivity and sensitivity,good recovery and reproducibility,low requirement of applied analysis and strong practicality,so it has good popularization value.Part II: Residues and Exposure Assessment of Tetracycline Antibiotics in Animal origin FoodObjective:Detection of Wuhan City and Hengyang City,animal-derived foods in the tetracycline antibiotic residues,which can account for the differences between the type and content of tetracycline antibiotics,,and through the national diet standards to assess its harm to the human body.Methods: The random sampling of Wuhan City and Hengyang City,the collection of markets and supermarkets in the animal food,through the first part of the method to detect the amount of tetracycline antibiotics residue,through the spss software on the two cities of tetracyclines Antibiotic content and species differences were analyzed.Combined with the dietary survey data of Chinese residents,the Monte Carlo method was used to simulate the level of tetracycline antibiotics exposed to food by Chinese residents,and to provide the basis for risk analysis and risk management.Result:(1)The detection rate of tetracycline antibiotics in animal origin food samples of Wuhan was higher than that of doxycycline(183/351)>chlortetracycline(56/351)> tetracycline(48/351)>(39/351),and the content of tetracycline antibiotics in central and far urban areas was statistically significant(P <0.05);meat,milk,aquatic and eggs were found in bazaars and supermarkets(P <0.05),the difference was statistically significant(P <0.05).(2)The detection rate of tetracycline antibiotics in animal food samples of Hengyang was higher than that of oxytetracycline(104/114)>tetracycline(91/114)> doxycycline(6/114)> chlortetracycline(4/114).The differences of meat,milk,aquatic products and eggs in the bazaars and supermarkets were statistically significant(P <0.05).(3)The difference of four kinds of tetracycline antibiotics between the two cities was statistically significant,and the detection rate of chlortetracycline and doxycycline was higher than that of Hengyang City,while oxytetracycline and tetracycline showed Hengyang City,the detection of higher than the detection level of Wuhan City;through the two cities of tetracycline antibiotic content distribution found: Wuhan City and Hengyang City,the difference between the four antibiotics were statistically significant,were expressed in Hengyang City Tetracycline content is higher than Wuhan City.(4)Spearman correlation analysis showed that the CTC with DOX,OTC and TC tetracycline antibiotics detection levels were positively correlated relationship between the components,and CTC and OTC,OTC and DOX,TC and DOX,tetracycline class antibiotics detection levels showed a negative correlation relationship between components.(5)Corresponding relations between different categories of whether bias pollution in food of animal origin of tetracycline antibiotics species differences.Conclusion: Tetracycline antibiotic exposure risk should not be ignored,affects antibiotics harmful factors on human health is of great significance to the early prediction and intervention measures.
