| Objective:The effect of An Wei Yu Yang Granule on GAS of Gastric Mucosa Injury Rat Induced by NSAIDs were studied and the protective effect of An Wei Yu Yang Granule on NSAIDs related gastric mucosal injury,and Investigate its pathogenesis were interpreted by rigorous,scientific animal experiment,so as to clinical treatment of this disease,to provide theoretical basis and objective guidance.Methods:Sixty healthy SPF level SD male rats adaptive feeding for 7 days,aged 2 month and weighted 180-220 g.The rats were randomly divided into six groups for ten rats in each group according to the random number table.A,B,C,D,E,F group respectively for negative control group,model group,rabeprazole group,higher-,medium-and lower-dose groups.A group and B group were given normal saline 1ml/100g/d,while C group was given,rabeprazole1.8mg/(kg/d)for intragastric administration.D,E,F groups were treated with higher-,medium-and lower-dose Of An Wei Yuyang Granule every day at 10 a.m,after 1 h,A group were given normal saline again,B,C,D,E and F group were given intragastric administration of diclofenac 9.89mg/kg/d,night fasting,for 7 consecutive days.All animals were anesthetized at the end of the last administration,and then laparotomized,exposure to the stomach and fixed.The mucosal blood flow was measured,and the blood was collected from the abdominal aorta,the content of GAS and MTL were prepared to measure.After laparotomy,cut the stomach along the greater curvature of the stomach and the gastric mucosa was observed by naked eyes generally,and then the stomach was fixed with 10% Formaldehyde Solution.Results:(1)Gastric mucosal blood flow(GMBF): The gastric mucosal blood flow level of model group were decreased compared with the control group,the other groups of rats' gastric mucosal blood flow were increased significantly,with statistical significance(P<0.01),in which an Wei Yuyang Granule high dose group increased most significantly;(2)Gastrin(GAS),Motilin(MTL): The gastrin and motilin level of model group significantly increased compared with the control group,with statistically significant(P<0.01),the other group of GAS levels were decreased,especially in the high dose group increased most significantly,with statistical significance(P<0.01);(3)The observation of light microscope pathology: Pathological results showed that the pathological changes of the gastric mucosa were especially severe in the model group.The control group and the traditional Chinese medicine group were mildly changed,and the high dose of Anwei Yuyang granule was the most light.Conclusion:?An Wei Yuyang Granule has a significant protective effect on gastric mucosal damage rat Induced by NSAIDs,and can increase the blood level of gastric mucosa,reduce the level of GAS and MTL;?Increased the level of GAS and MTL is an important factor in gastric mucosa injury caused by non steroidal drugs;?The prevention of An Wei Yuyang Granule on NSAIDs related gastric mucosal injury in rats may be through increased blood level of gastric mucosa,inhibit the expression of GAS,MTL to protect against oxygen free radicals,improve inflammation,thereby reducing gastric mucosal erosion and ulcer to achieve. |
