ObjectiveThis study aims to explore whether EGCG can improve diabetic complication myocardial fibrosis in intraperitoneal injection of streptozotocin rats through the TGF-?1/JNK signaling pathway.MethodsFifty male SD rats were divided into control group,model group,EGCG10 mg /kg,EGCG 20 mg / kg and EGCG 40 mg / kg.One-time intraperitoneal injection of STZ(65mg / kg),diabetic rat model were made.Each group of EGCG was given the above dosage,the control group and the model group were given 20 mg / kg of triple distilled water continuously for 12 weeks.The Glucose,Triglyceride,Cholesterol,Low Density Lipoprotein and High Density Lipoprotein were detected by ELISA in all the rats.Myocardial function was observed in diabetic cardiomyopathy rats by HE and Masson staining and Sirius red staining.Immunohistochemistry was used to detect the expression of Collagen?,Collagen?was detected to detect the degree of myocardial fibrosis in diabetic rats.Western blot was used to detect the expression of MMP-9,TIMP-1,TGF-?1,JNK and p-JNK in myocardium.ResultsCompared with the model group,the results showed that the serum glucose,triglyceride,cholesterol,and low-density lipoprotein of diabetic rats were decreased while the high-density lipoprotein of them was increased(P<0.05).Cardiac index and left ventricular mass index of the rats were decreased while heart function of them was improved(P<0.05).In addition,collagen I and collagen III proportional were regulated.The expression of TGF-?1,JNK,p-JNK,TIMP-1 were down-regulation and MMP-9 were up-regulation(P<0.05).ConclusionsEGCG can reduce the general biochemical indicators of diabetic rats,and improve the cardiac function and myocardial fibrosis in diabetic rats,and improve myocardial fibrosis in STZ-induced diabetic rats through TGF-?1/JNK signaling pathway.
