ObjectiveDetecting the expression of substance P(SP)and NK1 receptor(Neurokinin 1 receptor,NK1R)in eosinophils with allergic rhinitis(AR)and allergic rhinitis complicated with asthma(AR+AS)stimulated by different allergens,explore the role of SP and allergens in the pathogenesis of the disease.MethodsThe expression of SP and NK1R in peripheral blood eosinophils stimulated by Artemisia sieversiana wild allergen extract,dermatophagoides pteronyssinus allergen extract and Platanus pollen allergen extract in AR and AR+AS patients were detected by flow cytometry.The levels of SP and related cytokines level in plasma were detected by ELISA.ResultsPlasma SP levels in AR and AR+AS patients were 1.37 and 1.41 fold higher than healthy subjects,respectively,and positive correlated with plasma IL-4 and IL-17A levels.Compared with healthy control group,the ratio of SP and NK1R in peripheral blood eosinophils in AR and AR+AS patients were up-regulated.The Artemisia sieversiana wild allergen extract and Platanus pollen allergen extract could induce SP~+eosinophil increased expression.Artemisia sieversiana wild allergen extract,dermatophagoides pteronyssinus allergen extract and Platanus pollen allergen extract could promote the expression of NK1R~+eosinophils in AR patients,and the mean fluorescence intensity of NK1R on the surface of eosinophils of AR patients could be enhanced by Artemisia sieversiana wild allergen extract.There was no significant difference in the expression of SP and NK1R in healthy human eosinophils stimulated by different allergens.ConclusionThe level of plasma SP was increased in patients with AR and AR+AS,and the expression of SP and NK1R on eosinophils was up-regulated,allergen promotetheexpressionofSP~+eosinophils.Demonstratedthat eosinophil-derived SP and NK1R were play an important role in the pathogenesis of AR and AR+AS,allergens may be involved in the disease through the SP,SP and NK1R may be a new target for the treatment of AR and AR+AS.
