Objective:Depression has gradually developed into a common and high-incidence disease in modern society,it is commonly associated with gastrointestinal dysfunction.This study is based on the similarity of caused by depression and depression caused by "liver depression" in Chinese medicine.The change of NPY and leptin in the hypothalamus and gastric tissues of rats is based on the depression model,and the depression model of the Shuganhewei decoction is observed.The effects and mechanisms of rat feeding and gastric motility are the basis for further study on the pathogenesis of liver depression.Method:1.Theoretical study: first of all,the literature analysis method was adopted to sum up the understanding of " disorder of the stomach Qi " in Chinese medicine,and to comb the dission of the syndromes of estrus and gastric disappearance related diseases in the theory of “treatise on febrile and miscellaneous diseases”,and to analyze the relationship between NPY,leptin and depression,feeding and gastric motility,and analyze the phase of stomach depression caused by liver depression in Chinese medicine and the phase of NPY and leptin CUMStoms.2.The experimental study:(1)using chronic mild unpredictable stress improvement combined with soliton method,new feeding inhibition test was used to detect the change of feeding behavior and gastric emptying rate in the depression model rats.(2)Detection of related indexes: the relative expression of NPY,leptin,NPY1 R,NPY2R and OB-R in the gastric tissue of the depression model rats and the relative expression of NPY and leptin mRNA in the hypothalamus were detected by reverse transcription polymerase chain reaction;the fluorescence density of NPY1 R,NPY2R and OB-R in the depression model rats was detected by immunofluorescence.The expression of NPY and leptin in hypothalamus and gastric tissue of depression model rats was detected by ELISA.Results:1.Theoretical research: emotional factors can affect the symptoms of gastrointestinal dysfunction.Modern medicine believes that gastrointestinal dysfunction is associated with the hypothalamic feeding related signals and peripheral gastric motility,so we can explore the mechanism of "disorder of the stomach Qi" from the "liver depression" from the intake related signal NPY and leptin.2.Experimental study:Experiment 1: Effects of Shugan Hewei decoction on the ingestion inhibition experiment in depressed ratsCompared with the blank group,the incubation period of the model group was prolonged,the difference was statistically significant(P<0.01).Compared with the model group,the incubation period of the high dose group was significantly shorter than the model group(P<0.05).Compared with the fluoxetine group,there was no significant difference between the other treatment groups(P>0.05).Experiment 2: Effects of depression model rats on gastric emptying in depressive rats.Compared with the blank group,the gastric emptying rate in the model group was significantly lower,the difference was statistically significant(P<0.01),and the gastric emptying rate in the high dose group of Shugan Hewei decoction was higher than that in the model group(P<0.05),and the difference between the high dose group and the fluoxetine group was statistically significant(P<0.01).Experiment 3: effects of Shugan Hewei decoction on expression of NPY,leptin mRNA and protein in hypothalamus and stomach of rats with depression.Compared with the blank group,the relative expression of the gastric tissue and the hypothalamus NPY mRNA in the model group was significantly lower,and the difference was statistically significant(P< 0.01).Compared with the model group,the relative expression of NPY mRNA in the hypothalamus of Shuganhewei decoction and the high dose group increased,and the relative expression of NPY mRNA in the gastric tissue of Shuganhewei decoction was increased.The difference was statistically significant(P<0.01 or P<0.05);and the relative expression of NPY mRNA in gastric tissue in Shuganhewei decoction was higher than that in the fluoxetine group.The difference was statistically significant(P<0.01 or P<0.05).The content of NPY in the hypothalamus of the low dose group of Shugan Hewei decoction and the content of NPY in the gastric tissue in each dose group and the fluorine in each dose group There was no significant difference between the two groups(P>0.05).Compared with the blank group,the relative expression of gastric tissue and leptin mRNA in the hypothalamus increased significantly in the model group,and the difference was statistically significant(P<0.01).Compared with the model group,the relative expression of the gastric tissue and the hypothalamus leptin mRNA in each dose group of Shugan Hewei decoction and fluoxetine group decreased significantly(P<0.01).Compared with the fluoxetine group,the relative expression of the gastric tissue and the hypothalamus leptin mRNA in each dose group of Shugan Hewei decoction was significantly lower,the difference was statistically significant(P<0.01).The content of leptin in the hypothalamus and the content of leptin in the gastric tissue of the low dose group of Shuganhewei decoction and the content of leptin in the gastric tissue in each dose group were not statistically different from that of the fluoxetine group.(P>0.05).Experiment 4: effects of Shuganhewei decoction on expression of NPY1 R,NPY2R,OB-R and mRNA in gastric tissue of rats with depression.Compared with the blank group,the expression of NPY1 R in the model group was significantly reduced and the fluorescence density decreased,and the difference was statistically significant(P<0.01).Compared with the model group,the expression of NPY1 R in the high dose group of Shuganhewei decoction increased and the fluorescence density increased,the difference was statistically significant(P<0.01),and the expression of NPY1 R in the high dose group of Shuganhewei decoction was higher than that of the fluorine.The difference between the two groups was statistically significant(P<0.05).Compared with the blank group,the expression of NPY2 R in the model group increased significantly and the fluorescence density increased,and the difference was statistically significant(P<0.01).Compared with the model group,the expression of NPY2 R in each dose group of Shuganhewei decoction decreased and the fluorescence density decreased,the difference was statistically significant(P<0.01),and the expression of NPY2 R in each dose group of Shuganhewei decoction was lower than that of the model group.The difference between fluoxetine group and control group was statistically significant(P<0.05 or P<0.01).Compared with the blank group,the expression of OB-R in the model group increased significantly and the fluorescence density increased,and the difference was statistically significant(P<0.01).Compared with the model group,the expression of OB-R in Shuganhewei decoction and the high dose group decreased and the fluorescence density decreased,the difference was statistically significant(P<0.01),and the OB-R expression of the high dose group of Shuganhewei decoction was expressed in the model group(P<0.01).Compared with fluoxetine group,the difference was statistically significant(P<0.05).Conclusion:1.Emotions affect the liver's dispersal,and emotional depression and stomach drop are often treated by soothing the liver and stomach.2.Shuganhewei decoction can regulate the feeding behavior and gastric motility of the depression model rats,and can reverse the abnormal levels of mRNA and protein of NPY and leptin in the hypothalamus and gastric tissues of the model rats and the abnormal levels of mRNA and protein of NPY1 R,NPY2R,OB-R in the gastric tissue.It suggests that the liver and stomach soup may be through the up regulation of the hypothalamus and stomach tissues.NPY,NPY1 R and down regulation of leptin,NPY2 R and OB-R play an important role in regulating feeding behavior and improving gastric motility.3.The mechanism of "disorder of the stomach Qi" caused by liver depression in Chinese medicine may be related to the abnormal decrease of NPY and NPY1 R in the hypothalamus and gastric tissue and the abnormal rise of leptin,NPY2 R and OB-R.
