Objective The protective effect of tert-Butylhydroquinone?tBHQ?on the retina of type 2 diabetic rats induced by small doses of streptozotocin?STZ?and high-glucose-high-fat diet was investigated.The mi RNAs that were differentially expressed in rat retina after tBHQ intervention were identified.Finally,the role of differential miRNAs were analyzed by bioinformatics techniques to seek potential protection mechanisms of tBHQ,providing potential targets for prevention and treatment of DR.Methods 18 male SD rats were randomly divided into two groups:normal group and diabetic group.The type 2 diabetic rat model was established by high-glucose-high-fat diet feeding combined with small doses of STZ(30mg·kg-1)intraperitoneally.After established type 2 diabetic rats successfully,the rats further divided into diabetic group and tBHQ intervention group.The diabetic rats were sacrificed after intervention for 3 months to collect the fasting blood for measuring the biochemical and insulin-related indicators.HE staining was used to observe the pathological changes of retinal tissue.The mi RNA expression profile microarray was used to measure the differential miRNAs of rat retinas.The expression levels of specific miRNAs in the three groups of rat retinas were verified by PCR.The relevant pathways of differential miRNAs were analyzed.Results The fasting blood glucose in the tBHQ intervention group was higher than that in the normal group?P=0.00?,which was lower than that in the diabetic group?P=0.01?.The insulin in the tBHQ intervention group was lower than that in the normal group?P=0.00?,but it was higher than that in the diabetic group?P=0.00?.Severe retinal edema appeared in the retina of the diabetic group,and the structural layer was unclear based on HE staining.The pathological changes of the retina in the tBHQ intervention group were slight.Compared with diabetic rats,mi R-325-3p and miR-551b-3p were up-regulated?>1.5-fold?and miR-652-3p was down-regulated?>2.0-fold?in the tBHQ group.Mi R-325-3p and mi R-551b-3p were identified as differential miRNAs after PCR verification.By combining the results of the microarray experiments,we found that 21 pathways might be subject to miR-325-3p regulation from tBHQ treatment.Conclusion tBHQ has protective effects on diabetic retina through mi R-325-3p and its mediated 21 signaling pathways.
