| ObjectiveAlthough the diagnosis and treatment of colorectal cancer(CRC)have experienced constant innovations and development,the prognosis remains poor.The traditional CRC screening methods are less specific and sensitive.And colorectal endoscopy is an invasive examination with a certain risk.Therefore,there is an urgent need for a screening method that can detect early CRC patients,prolong the patient’s survival and improve the prognosis.Amino acids are the cornerstone of all life.Meanwhile,disease and health are directly or indirectly associated with amino acids.This study is based on blood samples of healthy people,colorectal adenoma and CRC patients,by liquid chromatography tandem mass spectrometry(LC-MS/MS),comparative analysis of differentially expressed metabolites of amino acids among the 3 groups.Screening markers can be used to predict early colorectal adenomas and CRC.Furthermore,the mechanism of CRC was revealed,providing a new direction for early diagnosis of CRC and drug therapyMethodsAfter an overnight fasting,3ml to 5mL of blood was collected among healthy individuals,colorectal adenoma and colorectal cancer patients respectively.The pretreatment of blood samples was carried out.And the differences of amino acid metabolism among 3 groups were analyzed byLC-MS/MS technique.ResultsThe amino acids and derivatives with significant differences between normal and colorectal adenomas are Et N,Tau,Hyp,Asp,Hcy,PEtN,Glu,Arg,Cit,PSer,Hcit,Trp,5HTP,Car,Asa,Orn,Lys,Hyl,Cth,Hpro,Sar,GABA,2Aib,5-Ava,Val,Hser,Leu,6Aca,Cys / Tau(P < 0.05).The amino acids and derivatives with significant difference between normal and colorectal adenocarcinoma are EtN,Tau,Hyp,Asp,Hcy,PEtN,Glu,Arg,Cit,PSer,Hcit,Trp,Orn,Lys,Hyl,Cth,Hpro,Sar,GABA,2Aib,Val,Hser,6Aca,Kyn / Tyr,Cys /Tau(P < 0.05).The amino acids and derivatives with significant difference between colorectal adenomas and colorectal adenocarcinoma are EtN,Cit,Val,Leu,Kyn / Tyr(P<0.05).ConclusionsAmino acids(Orn,Cit,Arg)involved in urea cycle,branched chain amino acids(Leu,Val),EtN and other amino acids,Kyn / Trp and Cys / Tau in colorectal cancer significantly changed.Amino acids occurred significantly change during in the development of colorectal cancer.Amino acids are potential biomarker in the early diagnosis of colorectal cancer. |
