ObjectiveTo observe the effect and mechanism of the Euonymusalat extract on nonalcoholic fatty liver fibrosis(nonalcoholic fatty liver fibrosis),and provide a theoretical basis for clinical practice.Methods72 clean SD rats,male,were randomly divided into polyene phosphatidylcholine group and ghosts and arrow feather extract group.To establish an animal model of nonalcoholic fatty liver fibrosis.At the end of the modeling,2 rats were randomly selected from each of the models,and the microscopic pathological changes were observed after microscopic examination.The standard of success was 30% of the hepatic steatosis in rats.The success of modeling was achieved.After the mold was finished,the medicine was administered the next day,and the drug was used for 3 weeks.The last time after intragastric administration,the removal of the eye blood,serum aspartate aminotransferase(aspartate,aminotransferase,AST)and alanine aminotransferase(alanine,aminotransferase,ALT),total bilirubin(TBIL)and serum HA and LN levels;observe the color and texture of the liver,liver tissue more than another poly Formaldehyde Solution fixed alternate for immunohistochemical detection;ultrastructural detection of liver from liver tissue in 5% glutaraldehyde solution.The content of malondialdehyde(malondialdehyde,MDA)in liver tissue was determined by thiobarbituric acid reactant colorimetric method,double antibody sandwich ELISA method was used to detect serum IL-6 and TNF-alpha levels,Western blot was used to detect TLR4 and HO-1 protein expression in rat liver tissue.The content of MMP-1 and JAK2 protein in liver tissue was detected by immunohistochemistry,and the expression of MMP-1 and JAK2 m RNA in liver tissues was detected by reverse transcription polymerase chain reaction(RT-PCR).ResultsIn the normal group,the general condition was good,the mental state and activity were all strong and the spirit was full.The spirit of model group rats gradually dispirited,slow,yellow urine,eating and appetite are poor,messy dull dark fur.The liver morphology of the rats in the high,middle and low dose groups of polyene phosphatidylcholine group and the extract of Euonymusalat improved in different degrees.The rats in the normal group had sharp edges,smooth surfaces and soft texture.The rats in the model group had blunt edges,smooth surfaces,tight membranes,uneven surface and rough surfaces,and oily faces.The liver morphology of rats in the extract group of polyene phosphatidylcholine group and high,medium and low dose group of euonymusalat improved.The normal group rats under light microscope cytoplasm stained red uniform nuclei normal morphology,nuclear stained;liver tissue of model group mice in tumor infiltrating lymphocytes,liver sinus silted up red blood cells and hyperchromatic nuclei and large patchy necrosis of liver cells,cytoplasm pale staining loose,lobular hyperplasia of fibrous tissue obviously.In the extract group of polyene phosphatidylcholine group and high,medium and low dose group of euonymusalat,liver tissue structure improved significantly;compared with the normal group,model group FD,HB,LIN,S were significantly increased(P < 0.05);compared with the model group,the extract group of polyene phosphatidylcholine group and high,medium and low dose group of euonymusalat score level was significantly decreased(P < 0.05);the normal group: rat liver stromal cells and nuclei were normal,the nucleus is located in the center of cell membrane is nearly circular,smooth.Model group: the rat capillary bile duct dilated,the cell volume of the hepatocyte increased,swelling,the activity of some enzymes on the membrane,and the swelling of the cytoplasm.The liver cell morphology of the rats of the extract group of polyene phosphatidylcholine group and high,medium and low dose group of euonymusalat of ghosts improved in different degrees.Compared with the normal group,model group rats serum ALT,AST and TBiL levels were significantly increased(P < 0.05);compared with the model group,the extract group of polyene phosphatidylcholine group and high,medium and low dose group of euonymusalat rats serum ALT,AST and TBiL were decreased(P < 0.05);compared with the normal group,the level of serum HA and LN rats the model was significantly increased(P < 0.05);compared with the model group,the extract group of polyene phosphatidylcholine group and high,medium and low dose group of euonymusalat rats serum HA,LN levels were significantly decreased(P < 0.05);compared with the normal group,significantly decreased the content of MDA in liver tissue of rats in model group(P < 0.05);compared with the model group,MDA significantly increased the content of the extract group of polyene phosphatidylcholine group and high,medium and low dose group of euonymusalat in liver of rats(P<0.05);compared with the normal group,model group rats serum IL-6,TNF-levels increased significantly(P<0.05);compared with the model group,the extract group of polyene phosphatidylcholine group and high,medium and low dose group of euonymusalat rats serum IL-6,TNF-levels decreased significantly(P<0.05);Compared with normal group,TLR4 increased significantly,the expression of HO-1 protein in liver tissue of rats in model group(P<0.05);compared with the model group,TLR4 were significantly decreased,HO-1 protein expression of the extract group of polyene phosphatidylcholine group and high,medium and low dose group of euonymusalat of rat liver tissue(P<0.05);compared with the normal group,the expression of MMP-1 protein in liver tissue of model rats decreased(P<0.05);compared with the normal group,the expression of JAK2 protein in liver tissue of model rats increased significantly(P<0.05);compared with the model group,the expression of the extract group of polyene phosphatidylcholine group and high,medium and low dose group of euonymusalat of liver JAK2 protein was significantly decreased(P<0.05);compared with the normal group,the expression model of liver group MMP-1 mRNA decreased significantly(P <0.05);compared with the model group,the expression of the extract group of polyene phosphatidylcholine group and high,medium and low dose group of euonymusalat MMP-1 in liver tissue of group mRNA were significantly increased(P <0.05);compared with the normal group,JAK2 mRNA in liver tissue of rats in the model group was significantly Increased(P<0.05),and compared with the model group,the expression of JAK2 mRNA in the liver tissue of the extract group of polyene phosphatidylcholine group and high,medium and low dose group of euonymusalat was significantly decreased(P<0.05).ConclusionEuonymusalat extract can improve liver inflammation,improve liver function,liver injury,inhibition of lipid peroxidation in liver,regulation signal pathway,liver cell structure and function to protect the integrity of fibrosis of liver fibrosis development trend,and in a dose dependent manner.
