Objective: Recently,nonalcoholic fatty liver disease(NAFLD)has become the most common cause of chronic liver disease with the increasing prevalence of obesity,diabetes,and the metabolic syndrome in the general population.Nevertheless,it still has no obvious treatment mechanism so far.Artesunate is the water soluble derivatives of artemisinin which shows high activity against both drug-resistant and drug-sensitive of malaria,it also has obvious anti-inflammatory effects.The maim of this study was to explore whether artesunate was able to improve NAFLD through TLR4/PI3K/Akt signal pathway.Methods: human hepatocyte LO-2 cells were treated with oleic acid(20 ?g/mL)for 48 hours,induces the NAFLD cell model and treated this model with different concentrations of artesunate,to see whether artesunate(Art)can reduce far degeneration,it has an effective protective effect on NAFLD.Detemined by MTT,Western blot,ELISA and immunofluorescence method of detection of oleic acid induced NAFLD cells survival and artesunate treatment of NAFLD in TLR4,PI3 K and Akt,the NF-?B,TNF-?,IL-10,IL-18 and IL-6 path expression changes and the changes of enzyme expression.Results: Artesunate reduces NAFLD-associated liver TG,ALT,AST and ALP contents(*p<0.05,#p<0.05),Artesunate could down-regulate the expression of TLR4/PI3K/Akt pathway and reduce the expression NF-?B transcription factor of NAFLD cells(*p<0.05,#p<0.05).Artesunate could reduce the content of inflammatory cytokines of NAFLD cells(*p<0.05,#p<0.05).Effect of artesunate on the oxidative response of NAFLD cells(*p<0.05,#p<0.05).Conclusion: Artesunate can improve the pathological status of NAFLD in a certain extent,it mainly works through the regulation of TLR4/PI3K/Akt signaling pathway,reducing the expression of inflammatory factors such as TNF-?,IL-6 and IL-18 and improvement of oxidative stress injury.
