Objective Sports fatigue is a physiological process that can not maintain a specific level in the process of exercise,it is an important factor to limit the effect of physical training.while acupuncture precounditioning therapy is both greenness and non-poisonous,if used properly,it can achieve good effect on preventing and treating sports fatigue.In this experiment,a rat model of exercise-induced fatigue was established by treadmill running,treated by acupuncture,taking energy metabolism of quadriceps femoris as the introducti-on,with observation on ATP levels,find the signs of fatigue in muscle as long as blood,also the microstructural structure of the four biceps femoris and the change of morphologic behavior in rat,in order to analysis the effect of "Acupuncture Preconditioning" on rats with exercise fatigue in detail,to explore the effect of "Acupuncture Preconditioning" on the prevention of sports fatigue in rats.Methods 40 male SD rats in this experiment,3-month-old,weighting 220±20g,provided by Hubei animal epidemic prevention center.The animals were kept in cages,with the freedom of feeding and drinking.Feeding room within constant temperature(22-24 degrees C)and humidity(55%),Daily replacement of padding,drinking water,cleaning drinking water,general feed total heat energy 3.8kcal/g(containing 70% carbohydrates,20% protein,10% fat).After two days of adaptive feeding,all rats were randomly divided into 4 groups,10 rats in each group.5 days of adaptability treadmill training except normal control group.Training program as follows: 10x10m/min on the first day,10x15m/min on second day,the third day is 18x15m/min,the fourth day is 21x20m/min,and the fifth day is 24x20m/min,than on the sixth days,these training rats were randomly divided into model group,acupuncture precounditioning group and post exercise acupuncture group according to body mass and exercise ability.Since then,40 mins is trained at a speed of 24 m/min every day,and the load intensity is equivalent to the maximum oxygen uptake of 60-70%.The whole process of training is 6 days a week,Sunday rest,acupuncture precounditioning group and post exercise acupuncture group respectively in each exercise 1 hours before and after acupuncture of ST36(Zu San Li),RN4(Guan Yuan),for 30 minutes,and operate 10 seconds each 5 minutes,the model group with no treatment for four weeks.The whole process of intervention in rats were observed and recorded the hair,urine,mental status;around intervention were observed quadriceps femoris tissue structure and mitochondrial ultrastructure,the detection injury markers in quadriceps femoris,serum markers of fatigue,and the ATP levels on mitochon-drial of quadriceps femoris.Results 1.Comparison of morphologic behavior of rats In the process of intervention,rats in model group appeared obvious rat hair sparse,loose stool symptoms,and the spirit in the training process of burnout,weakness,weakened after training;after acupuncture group, acupuncture precounditioning group showed slightly weak after training,the rest as usual.2.rat femoral head four in muscle tissue structure under electron microscope Microstructures under transmission electron microscope: the fasciculus of the rats' quadriceps femoris in the model group are irregular,loose,severely deformed and chaotic,the A brand disorder,I band twisted,H band is out of shape,there are also cracked Z line,the quantity of mitochondria decreased and with abnormal form.Both the two intervention group rats' quadriceps femoris fibre are partial rupture and disorder,just some of skeletal muscle cells swelling and vacuolization,A?I?H band distortion partial,only section mitochondria out of shape,others are still good.Less degree of damage in acupuncture preconditioning group than post exercise acupuncture group.3.Comparison of fatigue markers and blood serum metabolic markers in skeletal muscle of rats(1)MDA content of rat skeletal muscle(quadriceps femoris)and the contents of CK,BLA and BUN in serum: The intervention group was significantly lower than the model group(P<0.05)and significantly higher than that of the blank group(P<0.01),and there was a significant difference between the acupuncture precounditioning group and the model group(P<0.01),and the acupuncture precounditioning group was lower than that of the latter acupuncture group(P<0.05,P<0.01).(2)MG content and SOD activity of skeletal muscle:The intervention group was significantly higher than that of the model group(P<0.05),and significantly lower than that of the blank group(P<0.01),and there was a significant difference between the acupuncture precounditioning group and the model group(P<0.01),and the acupuncture precounditioning group was higher than that of post exercise acupuncture group(P<0.05,P<0.01). 4.Comparison of the ATP levels in the quadriceps femoris tissue of the rats The intervention group ATP levels of rats' quadriceps femoris tissue was significantly lower than the control group,with significant difference(P<0.01);The ATP levels of intervention group rats' quadriceps femoris tissue were significantly higher than those in the model group,there was significant difference(P<0.01),but the ATP levels of intervention group rats' quadriceps femoris tissue were lower than the normal control group,significant difference(P<0.01);the acupuncture preconditioning group rats' ATP levels of quadriceps femoris was higher than post exercise acupuncture group(P<0.05).Conclusion 1.In this experiment adopts treadmill running model,the experimental rats showed morphological behavior change of sports fatigue,and under the micro organization skeletal muscle injury,decreased serum MDA,BLA,CK,BUN,and increased the content of skeletal muscle MG,ATP content and SOD activity in skeletal muscle,suggesting that made the model of exercise-induced fatigue success.2.Acupuncture intervention can improve the structural morphological expression of quadriceps femoris in rats with exercise fatigue,improving the antioxidant capacity of skeletal muscle,decreasing the content of serum CK,BUN and LD,increasing the concentration of MG in skeletal muscle,reducing the content of MDA in skeletal muscle,enhancement of SOD activity in skeletal muscle,increasing the concentration of ATP in the mitochondria of skeletal muscle,reducing the structure and function of skeletal muscle mitochondria is affected,in order to prevent the occurrence of quadriceps femoris injury in sports fatigue. 3.Acupuncture preconditioning has more obvious advantages than post exercise acupuncture treatment in preventing and controlling sports fatigue.
