Objective: To research the expressions of B7-H3 and B7-H4 proteins which belong to the B7 protein family and the effect of sorafenib on their expressions in human hepatoma cell lines including HepG2,Hep3 B,BEL-7402,BEL-7404,BEL-7405,QGY-7701,QGY-7703,SMMC-7721,MHCC97 H,MHCC97L,HCCLM3 and HCCLM6.Methods:(1)We used Western blot(WB)to observe the expressions of B7-H3 and B7-H4 proteins in different human hepatoma cell lines(HepG2,Hep3 B,BEL-7402,BEL-7404,BEL-7405,QGY-7701,QGY-7703,SMMC-7721,MHCC97 H,MHCC97L,HCCLM3,HCCLM6)and normal human liver cell lines(HL-7702).The cell lines with high expression of B7-H3 protein and B7-H4 protein were screened.(2)After different doses of sorafenib were used,the MTT colorimetric assay and WB method were used to detect the expressions of B7-H3 and B7-H4 proteins in these selected cell lines.To further understand the inhibition of sorafenib on different human hepatoma cell lines and whether the overexpression of B7-H3 and B7-H4 proteins in different human hepatoma cell lines is common.(3)SPSS 21 statistics software(IBM,USA)and GraphPad Prism 5 were used to carry out statistical analysis and graphic display,and the measurement data were expressed as mean standard deviation(+ s).The data were compared and analyzed by one-way ANOVA,and then the differences between the different groups were further analyzed by post hoc analysis(Dunnett test).Results:(1)The expression of B7-H3 protein and B7-H4 protein in different human hepatoma cell lines was significantly higher than that of normal human hepatocyte cell line(HL-7702)(P<0.01).(2)The cytotoxic activity IC50 values of sorafenib on Hep3 B,BEL-7404,MHCC97 H,HCCLM3,and HCCLM6 with the most obvious overexpression of B7-H3 protein and B7-H4 protein were 14.56?M,9.14?M,9.46?M,17.21?M and 9.29?M.The expression of B7-H3 and B7-H4 in Hep3 B,BEL-7404,MHCC97 H,HCCLM3 and HCCLM6 liver cancer cell lines was significantly downregulated compared to the control group after 48 hours of treatment with sorafenib at doses of 5,10,and 20?M(P<0.01).Conclusion: The overexpressions of B7-H3 and B7-H4 proteins in different human hepatoma cell lines were common.Different doses of sorafenib could significantly reduce the expression of B7-H3 and B7-H4 protein in these 5 hepatoma cell lines.They might be regarded as candidate targets for prevention and treatment of hepatoma by affecting tumor immune evasion.
