Objective: To study the promoting effect of platelet-rich plasma(PRP)on the healing of lower limb tibial fracture in rabbits.Method:Sixteen healthy New Zealand white rabbit were selected and randomly divided into two groups.All rabbits were extracted with 5ml of blood before operation.All blood samples were centrifuged,and the plasma components were collected for detection.After the experimental group had collected blood samples,they were prepared for making of autologous rich platelet plasma.The same surgical method were performed in the fixation of all rabbits with posterior tibial fracture.Experimental group: autologous PRP group: autologous PRP extract was injected at the fracture site after surgery.Control group: normal saline was injected at the end of the fracture.Rabbit blood samples were collected at the day of surgery,2,4,8 and 12 weeks after surgery respectively,and the contents of Osteocalcin(OCN)and Bone Alkaline Phosphatase(BAP)in the blood were detected respectively.After the blood samples were collected,the rabbits were executed in batches of 2,4,8 and 12 weeks,and their lower limb fractures were treated with a general observation to evaluate the degree of fracture repair and the growth rate of Callus.Result:The PRP group,regardless of the diameter and volume of the callus in the initial stage,The difference of P=0.000(P<0.05)was statistically significant.The levels of osteocalcin in both groups peaked at week 4,and the difference between the two groups of P=0.000(P<0.05)was statistically significant.Bone alkaline phosphatase concentration,the peak of both groups appeared in week 2,P=0.000(P<0.05),and the difference of TW,TV% and blood vessel quantification statistically significant.At weeks 4,the difference between the two groups was statistically significant.Conclusion : Through animal experimental studies,it is shown that the autologous platelet plasma has an advantage in promoting fracture healing and repair.Its safety is high,and it's not easy to find hemolysis reation.It can provide a new way for orthopedic clinicians to treat fracture.
