The Expression Of Capn4 In Colorectal Cancer And Its Effect On Proliferation

Objective:To study the expression of Capn4 in the tissues of Colorectal cancer(CRC)and the corresponding colorectal tissue,and to explore the clinical significance of the three in colorectal cancer by combining the clinical and pathological parameters of Capn4 and colorectal cancer,and further explore its effect on the proliferation and apoptosis of colorectal cancer cells.Methods:From January 2011 to September 2016,the patients who were diagnosed as colorectal adenocarcinoma by biopsy of anterior bowel biopsy in the Second Affiliated Hospital of Nanchang University were used for this study.The tumor tissue and para colonic tissue were removed after the operation,and the cryopreservation was put into liquid nitrogen with the cryopreservation tube.The postoperative pathological results of the patients were screened before and after the screening.Pathological findings all suggested cryopreservation of colonic adenocarcinoma.A small block was taken to make a paraffin block in the hospital pathology department.The expression of Capn4 in CRC and corresponding para colonic tissue was detected by Immunohistochemistry(IHC).The remaining tissues extracted RNA,and two pairs of Capn4-specificationshRNA were synthesized by chemical synthesis method and transfected into colorectal cancer cells.The expression of Capn4 after transfection of SH RNA in colorectal cancer was detected by qRT-PCR and Western blot.The proliferation of colorectal cancer cells was detected by CCK-8.SPSS 19 software was used to analyze the experimental data.Results:1.Compared with adjacent tissues,the expression level of Capn4 in colorectal tumor tissues was significantly higher than that in adjacent tissues,and the expression difference was significantly statistical different(P<0.05).After that,western blot analysis showed that the higher expression of Capn4 protein was also found in tumor tissues.At the same time,the expression of Capn4 in CRC cells was also higher than that in normal human colorectal epithelial cell lines.2.Immunohistochemistry of human CRC samples(IHC)further confirmed the up regulation of Capn4 expression in CRC tissues.87(66%)positive CRC expression was evaluated in 132 samples,while weak positive staining was observed in non tumor tissues.These findings clearly indicate that Capn4 is overexpressed in CRC.3.by analyzing the clinicopathological data,the over expression of Capn4 is significantly correlated with the TNM stage and the size of tumor(P <0.05).In addition,the overall survival(OS)and disease-free survival(DFS)in the Capn4 high expression group were all lower than those in the low expression group(P <0.05).Conclusion:1.Capn4 is highly expressed in colorectal cancer tissues and cells.2.Capn4 overexpression is significantly correlated with TNM stage,tumor size and poor prognosis.3.Down regulating the expression of Capn4 significantly inhibited the proliferation of colorectal cancer cells.
