Part ?: Establishment of the model of endometrial adhesion in ratsIntrauterine Adhension(IUA)and Thin Endometrium(TE)are common causes of infertility.The application and popularization of hysteroscopic minimally invasive surgery can make the uterine cavity of the patients in the uterine cavity recover in different degree.However,although placing barrier system or biological agents in the intrauterine postoperatively has a certain effect in repair and maintenance of uterine cavity,adhesion postoperative is still common to occur,with infection risks.Especially in IUA of III-IV degree,the endometrium is extremely thin.And it has high postoperative recurrence rate,with low pregnancy rate and unsatisfactory reproductive prognosis.Only by finding effective ways to promote endometrial repair can the reproductive outcomes of these patients be improved.However,because of the difficulty of the administration routine and ethical reasons,it is urgent to find an available animal model.In this experiment,the aim is to build thin endometrium model and the model of intrauterine adhesions in rats,trying to bserve different dosing of gel or water which influence on rat endometrium recovery.Objective: to explore and construct the model of uterus adhesion and thin uterus in rats,and the effect of drug characters on therapeutic effect was also investigated.1)To establish an intrauterine adhesion model of rats,and provide animal model reference for the experimental study of endometrium.2)Explore the establishment method of thin endometrial model in rats;3)Fumble the drug delivery method in the uterine cavity of the rat,and provide a reference for the endometrial related experiment.4)To explore the drug delivery method of gelatinous drugs in the uterine cavity of rats,and to provide reference for the endometrial related experiment.Results:1)There was no significant difference in the thickness of endometrial model between the experimental group and the control group.2)There was a significant difference in the thickness of the endometrium between the experimental group and the control group,and the difference was statistically significant.There was a significant difference between the physiological saline group and the control group(P < 0.05),and the gel medium group was significantly different from the control group(P < 0.01).Conclusion:1)The method can effectively complete the modeling of the intrauterine adhesion model in rats with the triple damage method.2)The adhesion of rat uterine may be aggravated by the addition of gelatinous media after the modeling of rat uterine cavity adhesion model.Part ?: Expression and roles of Nuf2 gene in early Embryo development in MouseIt is estimated that the incidence of infertility worldwide is 8%-15%.And in China,nearly 10 million couples experience infertility.Declining fertility is becoming a great challenge to human health and reproduction.In 1978,the British scientist Edwards won the world's first successful in vitro fertilization embryo transfer(IVF-ET)by combining laboratory research with clinical medicine.In the past forty years,the number of children assisted by this technology and its derivative technologies has exceeded 6 million.It has become a large group.However,the success rate of ART is only about 30% at present,especially in the case of mature oocyte count,the live birth rate is below 5%,and the efficiency is very low.Because of the limitations of technology,methods and material sources,the complex mechanism of human early embryo development is not thorough enough.Therefore,understanding the mechanism of human early embryo development is very important.Studies have shown that Nuf2 is associated with the assembly of spindle and cell mitosis in the separation of cells.But no reports have been reported in the embryo.Objective: To explore the role of Nuf2 in mouse embryo development;Methods: Take embryos of 6 to 8 weeks ICR mice,inject Nuf2 Morpholino to inhibit Nuf2 protein expression.Then observing the phenotype of subsequent embryonic development process to determine whether Nuf2 gene is important or necessary in the process of embryonic development,in order to further explore its mechanism during embryo development.Result: After inhibiting the expression of Nuf2 protein,the mouse embryo was significantly inhibited during the development in vitro,and the rate of blastocyst and 4 cells /2 cells decreased significantly compared with the control group.The difference was statistically significant.Conclusion:Nuf2 plays an important role in early embryonic development of mice.
