The rapid and effective hemostasis is one of the important steps during surgical procedures.During the operation,the timely hemostasis in the wound could not only improve the possibility of the success,but also reduce the postoperative complications.With the development of science and technology,many more absorbable hemostatic agents was widely used,but these absorbable hemostatic agents only played the hemostatic effect,and these could not fill up the missing cells.So the wound would form the sags easily,and the healing time will be prolonged.Therefore,the hemostatic agent containing seed cells will be a new research direction.In this study,fibrinogen and thrombin were prepared for fibrin gel,to obtain a micro-environment with better hemostasis and cell compatibility.Then BMSC and vascular endothelial growth factor(VEGF)were loaded in the fibrin gel.Then induced the BMSC into the endothelial cells,to accelerate the healing and repair of the wound.The fibrinogen solution mixed with thrombin solution with a certain proportion,and 1:1,1:3 and 1:5 fibrin gels were successfully formed.The gels were semitransparent and tough.The swelling degree of 1:3 fibrin gel was found to be the largest.The result of HE staining and SEM showed that the gels were found to be a porous fiber network structure,and the pore size of 1:1 was similar to the 1:5,but both bigger than the 1:3.The result of procoagulant evaluation showed that the fibrin gel would activate platelets completely,and was very easy to form thrombus.The amount of thrombus formed on the 1:5 fibrin gel surface was the most and the time of APTT was prolonged.The result of suckling rat wound experiment showed that the 1:5 fibrin gel would promote the healing of the wound.The result of cell compatibility evaluation on fibrin gel surface showed that the fibrin gel surface was beneficial to the adhesion and proliferation of EC and SMC.The morphologies of EC and SMC were normal,and the activities and the proliferation rates of EC and SMC were higher than the blank.1:3 and 1:5 were superior than 1:1,but there was no difference between 1:3 and 1:5.The result of BMSC loaded in the fibrin gel showed that this fibrin gel could encapsulate BMSC,and could achieve uniform distributed of BMSC within the fibrin gels.BMSC in the fibrin gel appeared with cell division and spreading,and the cells showed long spindle shape or triangle.The cell activity of BMSC in 1:1,1:3 and 1:5 fibrin gel remained stable,and there were no difference among 1:1,1:3 and 1:5.The results of induced differentiation of BMSC embedded in fibrin gel showed that the growth of BMSC in fibrin gel was good,but the morphology of cell was found.The proliferation of BMSC was found,and the growth law of cell activity of the experimental group was similar to the control group.The effect of the experimental group on the differentiation of BMSC to endothelial cell was better than the experimental group.Above all,this study has successfully prepared a biological material with hemostasis and accelerating the rapid repair of the wound.The material has good hemostatic and coagulating effect,and the cell compatibility is good.This material could embed the cells,then the cells are used to repair the wound.This material provides a new solution for the large trauma surface.
