Objective: To make a stable fibrin glue in vitro,and investigate the effects of different concentrations of fibrinogen,aprotinin and tranexamic acid on dissolution rate of fibrin glue.Methods:Fibrinogen,thrombim,calcium chloride,aprotinin or tranexamic acid were used to make the different characteristics of fibrin glues.Fibrinogen was divided into three groups : group A(10 mg/ml),group B(15mg/ml),and group C(20mg/ml).Aprotinin was divided into three groups,containning group 1(15000KIU/ml),group 2(20000 KIU/ml),and group 3(25000 KIU/ml).Tranexamic acid was also divided into three groups,containning group?(10 mg/ml),group ?(30mg/ml),and group ?(50mg/ml).Group 0 without inhibitors was set as the control group.15mg/ml fibrinogen,25000 KIU/ml aprotinin,and 50mg/ml tranexamic acid were used to make the fibrin glue,which was marked as group U.Each group was placed in the 37? thermostatic water bath,with a close observation in the dissolved state.Results:(1)The milky glue like solid were formatted after 40 to 60 seconds.(2)The dissolved time was different in each group as follow: group A0,group A1,group A2,group A3,group A?,group A?,and group A? lasted for 4,11,13,13,4,7,and 8 days respectively;group B0,groupB1,group B2,group B3,group B?,group B?,and group B? lasted for 5,13,18,19,7,8,and 9 days;and group C0,group C1,group C2,group C3,group C?,group C?,and group C? lasted for 6,18,24,25,9,9,and 10 days.In a same concentration of aprotinin or tranexamic acid,the dissolute rate was significantly lower in the high dose of fibrinogen than that in the low dose of fibrinogen(P<0.05).In a same concentration of fibrinogen,when aprotinin was less than 20000 KIU/ml,the dissolute rate was lower in a higher concentration of aprotinin in a low concentration of aprotinin(P<0.05);but no significant difference in dissolute rate between different concentration of aprotinin when aprotinin was 25000 KIU/ml(P>0.05).In the same concentration of fibrinogen,the higher concentration of tranexamic acid slowed down the dissolved time(P<0.05).(3)The dissolution time in group B3,group B?,and group U lasted for 20,9 and 31 days.Compared with the single use of aprotinin or tranexamic acid,the dissolute rate was significantly lower in the aprotinin and tranexamic acid(P<0.05).Conclusion:(1)Higher concentration of fibrinogen,aprotinin,tranexamic acid could strengthen the stability of fibrin glue.The glue dissolution rate was inversely correlated with the concentration of fibrinogen and tranexamic.Aprotinin protecting glue from solution was better than tranexamic acid,when the concentration of aprotinin was less than 20000 KIU/ml,The glue dissolution rate was inversely correlated with the concentration of aprotinin,when the concentration of aprotinin was 25000 KIU/ml,there was no significant difference in glue dissolution rate.(2)The protective effect again glue solution with combination of aprotinin and tranexamic acid was significant superior to aprotinin or tranexamic acid alone.
