Primary liver cancer is a major disease that endangers human health,the fifth highest in malignancies,and the third in cancer.At present,to effect a radical cure of liver cancer are mainly clinical surgical resection and liver transplantation,but its low risk big,the removal rate,high recurrence rate become the biggest obstacle to treatment of liver cancer.The results showed that the recurrence rate of the radical resection of hepatocellular carcinoma was 80.0%,and the recurrence rate was 40.0% to 50.0%.Some small metastases due to the complexity of local anatomy of the liver,often hidden in the connective tissue,closely associated with important blood vessels around the viscera,intraoperative lymph node cleaning often incomplete,with blindness,lead to increased postoperative tumor recurrence,and liver malignant tumors grow more invasive and centricity,some patients already exists in the early years of the onset of cancer metastasis and local lymph node metastasis.For middle-late stage is not suitable for surgery patients mostly choose to chemotherapy,radiation therapy,interventional measures,such as,chemotherapy drugs on inhibiting tumor growth at the same time,also have damage to the body's normal tissues.How to improve the liver targeting effect on chemotherapy drugs,drug concentration in maintaining metastasis lymph nodes,reduce side effects to normal tissues become the liver cancer chemotherapy research and the focus of attention.Chemotherapy drug nanoparticles is a new type of directional controlled release method,by the nature of the nanometer carrier carrying the chemotherapy drugs and control drug sustained release in the target cell,form a high local concentration,within the metastases tumor cells to kill your inhibition at the same time,and to reduce the chemotherapy drugs brought about by the adverse reaction.In recent years,drug-loading nanoparticlestechnology because of its many advantages,has become a more cancers intraoperative tracer and targeted therapy research hot spot,but for liver cancer metastasis lymph nodes of the tracer and targeted chemotherapy research is less.Nano carbon is a kind of average diameter of 150 nm activated carbon particles,as a carrier of intraoperative tracer and passive targeting has been successfully applied to the current type of surgical treatment of the tumor,its mechanism is to use the carbon nanoparticles lymphatic tropism,super adsorption,and controlling functions carrying anticancer drugs after local injections,consumed by macrophages into the lymphatic system,and accumulated in the draining lymph nodes,to slow release chemotherapy drugs to extend the duration,and metastases in the cancer cells to kill or inhibit sustainability and also can reduce the dosage of enter blood circulation,reduce the adverse reaction.5-fluorouracil is the choice of the digestive tract tumor chemotherapy drugs,its cell RNA interference through and thymine DNA nucleotides synthetase and inhibiting tumor DNA synthesis,has the role of inducing tumor cell apoptosis.This experiment to establish the rat model of liver cancer,first set up two experimental group and control group,experimental group with nano carbon5-Fu mixed suspension carcinoma submucosal injection,control group 1 pure nano carbon mixed suspension with carcinoma submucosal injection,control group 2 with simple 5-Fu aqueous carcinoma submucosal injection,the six phase at the same time point and take its blood plasma cleaning the lymph nodes,using high performance liquid chromatography(HPLC)method,the test group and control group 2 5-Fu drug concentrations in the blood and lymph nodes,and calculate the selective index(SI)and targeting index(TI),with SI and TI nano carbon 5-Fu mixed suspension on rat liver cancer of the lymph node targeted,indirectly speculate anti-cancer effect,for further research on liver cancer patients with lymphatic chemotherapy to provide experimental basis.Objective: the nanometer carbon pick up 5-fluorouracil(5-Fu)made of suspension liquid is applied to the rat model of liver cancer,explore nano carbon 5-Fu mixed suspension effect on rat liver cancer lymph node of tracer,detection of chemotherapy drugs at different time points 5-Fu in the drug concentration in the lymph nodes,nano carbon 5-Fu suspension liquid and5-Fu aqueous solution on rat liver cancer lymph node,targeted for further research on liver cancer patients targeted chemotherapy to provide experimental basis.Methods: a total of 108 rats were randomly divided into three groups of36.Experimental group with nano carbon 5-Fu mixed suspension carcinoma subcapsular injection,control group 1 pure nano carbon mixed suspension with carcinoma subcapsular injection,control group 2 with pure aqueous solution 5-Fu carcinoma subcapsular injection,,30,60,120,240,480 15 min after the injection of each executed 6 rats in each group respectively,take the plasma and cleaning the lymph node:1 Compare the number of lymph nodes in the experimental group and the control group of 1 group,and examine whether the chemotherapeutic drugs were affected by the 5-Fu.2 Using high performance liquid chromatography(HPLC)method,test group and control group 2 5-Fu drug concentrations in the blood and lymph nodes,and calculate the selective index(SI)and targeting index(TI),with SI and TI nano carbon 5-Fu mixed suspension on rat liver cancer of the lymph node targeted.3 Adopt SPSS 17.0 software for statistical analysis.According to the data of the three sets of quantitative data,the comparison between the group and the independent sample T was analyzed,and the difference in P<0.05 was statistically significant.Results:1 The difference in the number of lymph nodes in the rats and the control group was statistically significant when the 5-Fu suspension was given under the pericardial periclum;2 The concentrations of 5-Fu in rats were significantly higher than those in the two groups,and the 5-Fu concentrations were significantly lower than those in the control group.And the experimental group was more than 1 in six different target index(TI)and selectivity index(SI).Conclusion: Nano carbon pick up after the chemotherapy drug 5-Fu does not affect its tracer,nano carbon 5-Fu mixed suspension of rat liver cancer lymphatic organization has good targeting property,can be a long time to maintain drainage lymph nodes in the chemotherapy drug concentration,at the same time can reduce the effective distribution of 5-Fu in peripheral blood,nano carbon is an ideal carrier in the rat liver cancer lymphatic chemotherapy.
