Study On Chemical Constituents And Bioactivities Of Aconitum Brevicalcaratum(Finet Et Gagnep.) Diels

The Aconitum family is an important medicinal plant in the folk.It has been used to cure pain,arrhythmic,tumor,Alzheimer's disease and control insects for thousands of years.It is found that diterpenoid alkaloids is the characteristic component and the main active components of the Aconitum family.Aconitum brevicalcaratum?Finet et Gagnep.?Diels,a member of the Ranunculaceae Aconitum family,widely distributed in northwestern Yunnan Province.Compared with other plants,it shows special characteristics for some diseases,such as rheumatism lumbocrural pain,antipyretic cough,sore throat and aconitine poisoning disease.However there is rarely reported about Aconitum brevicalcaratum?Finet et Gagnep.?Diels at present.In this work,the chemical composition and the biological activities of extraction with CH₂Cl₂ were studied systematically for the finding of novel drug from plant.The detail was described below.In this paper,The shade dried and powdered Aconitum brevicalcaratum?Finet et Gagnep.?Diels plant?5 kg?was soaked with EtOH.After removing the solvent,the ethanol extract was recovered and extracted by using acid extraction and alkali precipitation to gain crude.Thirteen coumpounds were isolated by methods of column chromatography and recrystallization.Their structures were elucidated on the basis of extensive spectroscopic analyses and document literature and comparison analysis.Among them,six new compounds were determined as Brevicanine I?1?,Brevicanine II?2?,Brevicanine III?3?,Brevicanine IV?4?,Brevicanine V?5?,Brevicanine VI?6?.Seven known compounds were determined as Scaconitine?7?,Acobretine C?8?,Acobretine D?9?,8-O-ethylscaconine?10?,methyl N-acetylanthranilate?11?,2-?acetylamino?-N-?2-methoxyphenyl?benzamide?12?,2-?2-methyl-4-oxo-4H-quinazoline-3-yl?benzoic acid methyl ester?13?.The cytotoxicity of compound 1-4 was investigated against SH-SY5Y,MCF-7 and HepG2 cell line,and the cytotoxicity of compound 7-10 was investigated against HCT116human cancer cell line,which showed none of them possessing considerable anti-proliferative activities;The biological activities of the chemical composition were studied.The effect of survival rate about compound 1-4 was investigated against SH-SY5Y cell under MPP⁺exposure,which showed Brevicanine II?2?has an protective effect;To evaluate the autophagy effect of compound 7-10,Western Blot was used to detect the expression of autophagic marker by stimulating human cancer HCT116 cells.The results showed that compound 7 induced protective autophagy in HCT116 cells.
